Vi beskriver en simpel og pålidelig metode til indsamling af sæd fra små og mellemstore fugle i reproduktivt aktiv tilstand, og uddrager DNA fra aviær sperm.

Fugle er ideelle temaer for undersøgelser, hvor sædkvalitet og konkurrence1, sperm telomere dynamics2, Epigenetik3og lignende emner, som de er meget udbredt i biologisk forskning og sperm nemt kan udtages ved hjælp af cloacal massage. Cloacal massage er en blid, ikke-invasiv manuel teknik til opsamling af sæd fra fugle4,5,6. Gentagne prøver kan opnås nemt og ingen særlige værktøjer er nødvendige, hvilket gør det nemt at udføre i det felt eller lab. Selv om cloacal massage har været anvendt i årtier, er det svært at lære fra de tilgængelige skrevet beskrivelser. Denne publikation er beregnet til at reducere den tid og usikkerhed, der er involveret i at lære cloacal massage.
Sæd opsamlet fra fugle ved hjælp af cloacal massage kan bruges straks til motilitet vurdering4,7 eller kunstig befrugtning8eller frosset til andre anvendelsesformål såsom avanceret billedbehandling og DNA-ekstraktion. Sæd prøver fra tyk fugle er små men indeholder tætpakkede sædceller. DNA er udvundet ved hjælp af en kommerciel udvinding kit for enkelhed, med ændringer til at overvinde de beskyttende specialfunktioner sperm9. Efter ekstraktion, kan sperm telomerer måles ved hjælp af qPCR10.

Opsamling af sæd kan være nyttige for en bred vifte af applikationer, herunder undersøgelser, hvor sædkvalitet, sperm telomere dynamik og Epigenetik. Cloacal massage og DNA udvinding ved hjælp af den beskrevne protokol er blevet udført på flere tyk arter og en columbid (tabel 1).
