Cloacal massasje er en mild, ikke-invasiv manuell teknikk for innsamling sæd fugler4,5,6. Gjentatt prøvene kan hentes lett spesial verktøy er nødvendig, noe som gjør det enkelt å utføre i feltet eller laboratorium. Selv cloacal massasje har vært i bruk i flere tiår, er det vanskelig å lære fra tilgjengelig skrevet beskrivelser. Denne publikasjonen er ment å redusere tid og usikkerhet involvert i å lære cloacal massasje.
Sæd fra fugler bruke cloacal massasje kan brukes umiddelbart for motilitet vurdering4,7 eller kunstig insemination8eller frosset for andre bruksområder som avansert bildebehandling og DNA utvinning. Sæd prøver fra ei gruppe fugler er små, men inneholder tettpakkede sperm. Samling av sæd kan være nyttig for en rekke applikasjoner inkludert studier med kvaliteten, sperm telomere dynamics og epigenetics. Cloacal massasje og DNA utvinning bruker beskrevet protokollen er utført på flere ei gruppe arter og en columbid (tabell 1).
