I denne artikkelen beskriver vi en helhjertet MR-protokoll for å avgrense endringer i eksperimentell cerebral malaria. Malaria er et betydelig globalt helseproblem. 1 Alvorlig malaria er preget av cerebral involvering og er ofte en dårlig prognostisk faktor. Cerebral involvering er vanlig hos barn under fem år i områder med høy malariatransmisjon og representerer den viktigste årsaken til malaria relatert død i denne aldersgruppen. 1 Mens aggressiv behandling kan være livreddende, kan det være vanskelig å påvise cerebral malaria, spesielt i tidlige stadier. De patologiske prosessene som er involvert i cerebral malaria inkluderer mikrovaskulær forstyrrelse og cerebralt ødem, noe som kan føre til alvorlig hjernesvulst. I denne artikkelen presenterer vi en magnetisk resonans imaging (MRI) protokoll som tillater hel hjerne in vivo avbildning av eksperimentell cerebral malaria (ECM). Hele hjernens høyoppløselige billeddannelsesmetoder har blitt mye underutilisert i denne sykdommen, selv om lite er kjent om hvordan ECM initierer i det sentraleNervesystemet eller hvilke spesifikke mekanismer som fører til sykdommen. Cerebral malaria is a sign of severe malarial disease and is often a harbinger of death. While aggressive management can be life-saving, the detection of cerebral malaria can be difficult. We present an experimental mouse model of cerebral malaria that shares multiple features of the human disease, including edema and microvascular pathology. Using magnetic resonance imaging (MRI), we can detect and track the blood-brain barrier disruption, edema development, and subsequent brain swelling. We describe multiple MRI techniques that can visualize these pertinent pathological changes. Thus, we show that MRI represents a valuable tool to visualize and track pathological changes, such as edema, brain swelling, and microvascular pathology, in vivo.
