Ondanks de voordelen van makkelijke toepasselijkheid en kosteneffectiviteit, hebben subcutane muismodellen ernstige beperkingen en worden de tumorbiologie en tumorcel-verspreiding niet nauwkeurig gesimuleerd. Orthotopische muismodellen zijn geïntroduceerd om deze beperkingen te overwinnen; Dergelijke modellen zijn echter technisch veeleisend, vooral in holle organen zoals de dikke darm. Om uniforme tumoren te produceren die op betrouwbare wijze groeien en metastaseren, zijn gestandaardiseerde technieken van tumorcelbereiding en injectie kritisch. We hebben een orthotopisch muismodel van colorectale kanker (CRC) ontwikkeld die zeer uniforme tumoren ontwikkelt en kan gebruikt worden voor tumorbiologie studies en therapeutische proeven. Tumorcellen uit primaire tumoren, 2-dimensionale (2D) cellijnen of 3-dimensionale (3D) organoïden worden geïnjecteerd in de cecum en, afhankelijk van het metastatische potentieel van de geïnjecteerde tumorcellen, vormen zeer metastatische tumoren. Daarnaast,CTC&#39;s kunnen regelmatig worden gevonden. Hier beschrijven we de techniek van tumorcelbereiding uit zowel 2D-cellijnen als 3D-organoïden, evenals primair tumorweefsel, de chirurgische en injectietechnieken, alsmede de isolatie van CTC&#39;s uit de tumorhoudende muizen en de huidige tips voor het oplossen van problemen.

De succesvolle en reproduceerbare generatie van colorectale tumoren in dit model hangt kritisch af van de nauwkeurige injectie van de cellen zonder spleten of lekkage. Als dit bereikt wordt, is dit model extreem betrouwbaar en resulteert het zeer zelden in kunstmatige peritoneale verspreiding.
