Fluen medulla neuropil er organiseret som et 3D gitter af 10 lag og cirka 750 kolonner. Først viser vi en dual-view billeddannelse metode, som bruger tilpasset software til at kombinere to konfokal billede stakke at generere høj kvalitet isotrope billeder. Arbejdsgangen indeholder tre afsnit: (i) dual-view imaging sektion kombinerer to konfokale billedstakke indsamlet på ortogonale orienteringer i en høj opløsning 3D billede af dendritter; (Ii) dendritceller sporing og sektion registrering spor dendritiskedorne i 3D og registrerer dendritiske spor til henvisningen kolonne matrix; (Iii) den dendritiske analyse afsnit analyseres dendritiske mønstre med hensyn til kolonner og lag, herunder lag-specifikke terminering og plane projektion retning af dendritiske dorne, og udleder estimater af dendritiske forgrening og terminering frekvenser. Protokollerne udnytte brugerdefinerede plugins bygget på open source MIPAV (Medical Imaging behandling, analyse og visualisering) platform og brugerdefinerede værktøjskasser i matricen laboratorium sprog. Tilsammen udgør disse protokoller giver et komplet workflow til at analysere den dendritiske routing af Drosophila medulla neuroner i lag og kolonner, for at identificere celletyper, og til at bestemme defekter i mutanter.

Brug af dual-view imaging procedure præsenteres her, blev en flue hjerne indeholder tyndt mærket Tm20 neuroner afbildet i to ortogonale retninger. Forud for billeddannelse, blev hjernen farves med passende primære og sekundære antistoffer til visualisering af membran-tøjret GFP og fotoreceptor axoner.
