Hier laten we zien hoe beeld en analyseren van dendritische priëlen van Drosophila merg neuronen. Tijdens de ontwikkeling neuronen uitgebreide dendrieten in complex maar stereotype vertakte patronen synapsen met hun presynaptische partners. Dendritische vertakking patronen correleert met neuronale identiteit en functies. De locaties van dendritische priëlen bepalen het type presynaptische inputs die zij ontvangen, terwijl de dendritische vertakking complexiteit en veld maten regelt de input nummer. Zo dendritische morfologische eigenschappen zijn cruciaal determinanten van synaptische connectiviteit en neuronale berekening. In veel regio&#39;s van complexe hersenen, zoals de vlieg optische kwabben en gewervelde netvlies worden synaptische circuits georganiseerd in lagen en kolommen informatieverwerking 1, 2 vergemakkelijken. In dergelijke kolom en laag organisatie presynaptische neuronen van een afzonderlijke modaliteit project axonen te eindigen bij een bepaalde laag (zogenaamde layer-specifieke targeting) en een ordelijke tweedimensionale matrix vormen (zogenaamde called topografische kaart), terwijl de postsynaptische neuronen uit te breiden dendrieten van de juiste afmetingen in bepaalde lagen om presynaptische ingangen van de juiste soorten en aantallen ontvangen. Terwijl axonale targeting lagen en kolommen is goed bestudeerd 3, 4, is veel minder bekend over hoe dendrieten worden naar specifieke lagen en vergroten de juiste maat receptieve velden synaptische verbindingen met de juiste partners presynaptische 5 vormen. De moeilijkheid van de beeldvorming en kwantificeren van dendritische richten naar lagen en kolommen heeft de studie van de dendritische ontwikkeling in zuilvormige en gelamineerde hersenstructuren belemmerd. Drosophila merg neuronen zijn een ideaal model voor het bestuderen van dendritische routing en circuit assemblage in de kolommen en lagen. De fly merg neuropil is georganiseerd als een 3D rooster van 10 lagen en ongeveer 750 kolommen. Elke kolom wordt geprikkeld door een set afferentia, waaronder photoreceptors R7 / R8 en lamina neuronen L1 - L5, waarvan de axonen terminals vormen topografische kaarten in een laag-specifieke wijze 6. Ongeveer 38 soorten merg neuronen zijn aanwezig in elke kolom merg en uitgebreide dendrieten in specifieke lagen en met de juiste veld maten om input te ontvangen van deze afferenten 7. De synaptische circuits in de medulla werden gereconstrueerd op het elektronenmicroscopische niveau; Zo zijn de synaptische samenwerking gevestigde 7, 8. Bovendien genetische hulpmiddelen voor etikettering verschillende soorten neuronen medulla zijn 9, 10, 11. Bij de behandeling van drie soorten transmedulla (Tm) neuronen (Tm2, Tm9 en TM20), hebben we eerder geïdentificeerd twee cel-type-specifieke dendritische attributen: (i) Tm neuronen projecteren dendrieten in zowel de voorste of achterste richting (vlakke projectie richting), afhankelijk van het type cel en (ii) dendrieten van neuronen medulla beëindigen specifieke medulla lagen in een celtype-specifieke wijze (laagspecifieke beëindiging) 12. Planar projectierichting en laagspecifieke beëindiging voldoende om deze drie soorten Tm neuronen differentiëren, terwijl mutaties die Tm reacties op laag en kolom signalen verstoren invloed verschillende aspecten van deze kenmerken. Hier presenteren we een complete workflow voor de behandeling van dendritische patroonvorming van Drosophila medulla neuronen in lagen en kolommen (figuur 1). Ten eerste tonen we een dual-beeldvorming methode, die aangepaste software gebruikt twee confocale beeld combineren stapels van hoge kwaliteit isotrope beelden genereren. Deze methode vereist slechts conventionele confocale microscopie om beelden van hoge kwaliteit die het mogelijk maken voor een betrouwbare tracering van dendritische filialen te genereren, zonder toevlucht te nemen tot super-resolutie microscopie, zoals eens STED (gestimuleerde emissie uitputting) of structurele verlichting. Ten tweede presenteren we een methode voor het opsporen dendritische assen en voor het registreren van de resulterende neurieten sporen een referentie kolommatrix. Ten derde, tonen we de computationele werkwijzen voor extraheren van informatie over het vlakke projectierichting en laagspecifieke beëindiging van dendrieten, en voor het afleiden schattingen dendritische vertakking en beëindiging frequenties. Samen zijn deze methoden is het mogelijk voor de karakterisering van dendritische patronen in 3D, de indeling van celtypen op basis van dendritische morfologie, en de identificatie van mogelijke gebreken in mutanten.

Dit rapport beschrijft de procedures voor het imago en de dendrieten van merg neuronen te analyseren. De workflow bestaat uit drie onderdelen: (i) de dual-view beeldvorming sectie combineert twee confocale image stacks verzameld op orthogonale oriëntaties een hoge resolutie 3D-beeld van dendrieten in; (Ii) de dendrite opsporing en registratie sectie sporen dendritischepriëlen in 3D en registreert dendritische sporen met de referentie-kolom-array; (Iii) de dendritische analyse deel wordt dendritische patronen ten opzichte van kolommen en lagen, waaronder laagspecifieke beëindiging en vlakke projectierichting van dendritische spillen afleidt ramingen van dendritische vertakking en beëindiging frequenties. De protocollen maken gebruik van aangepaste plugins gebouwd op het open-source MIPAV (Medical Imaging verwerking, analyse en visualisatie) platform en aangepaste gereedschapskisten in de matrix laboratorium taal. Samen vormen deze protocollen een volledige workflow de dendritische routering van Drosophila medulla neuronen in lagen en kolommen analyseren, om celtypen te identificeren, en om defecten in mutanten bepalen.

Met behulp van de dual-view beeldvorming procedure die hier gepresenteerd, een vlieg hersenen met dun gelabeld TM20 neuronen werd in beeld gebracht in twee orthogonale richtingen.
