Her viser vi hvordan du bildet og analysere dendrittiske lysthus av Drosophila medulla nerveceller. Den første delen, dual-view bildebehandling, beskriver deconvolution og kombinasjon av to bildestakker inn et bilde med høy oppløsning stabelen. Under utviklingen til nevroner forseggjort dendritter i komplekse, men stereotype forgrenede mønstre danner synapser med sine presynaptiske partnere. Dendrittiske forgrening mønstre korrelerer med nevronale identitet og funksjoner. Lokaliseringen av dendrittiske arbors bestemme hvilken type presynaptiske innganger de mottar, mens dendrittiske forgrening kompleksitet og feltstørrelser styrer inngangsnummer. Dermed dendrittiske morfologiske egenskaper er kritiske determinants for synaptiske tilkobling og nevronale beregning. I mange deler av komplekse hjerner, slik som fly optiske fliker og virveldyr hinnen, blir synaptiske kretser organisert i kolonner og lag for å lette informasjonsbehandling 1, 2. I en slik kolonne og lag organisasjon, til presynaptiske nerveceller i en distinkt modalitet prosjekt aksoner avsluttes ved et bestemt lag (såkalt lag spesifikk målretting) og til å danne en ordnet to-dimensjonal matrise (så-called topografisk kart), mens postsynaptiske nevroner utvide dendritter av passende størrelse i bestemte lag for å motta presynaptiske inngangene de riktige typene og tall. Mens axonal målretting til lag og kolonner har blitt godt undersøkt 3, 4, er langt mindre kjent om hvordan dendritter blir rutet til bestemte lag og utvide riktig størrelse mottakelig felt for dannelse av synaptiske forbindelser med de riktige presynaptiske partnerne 5. Vanskeligheten med bildebehandling og kvantifisere dendrittiske målretting til lag og kolonner har hindret studiet av dendrittiske utvikling i søyle og laminerte strukturer i hjernen. Drosophila medulla nevroner er en ideell modell for å studere dendrittiske ruting og kretsstevne i kolonner og lag. Flua medulla neuropil er organisert som en 3D-gitter av 10 lag og ca 750 kolonner. Hver kolonne er innervert av et sett av afferente, inkludert photoreceptors R7 / R8 og lamina nevroner L1 - L5, hvis aksonal terminaler danne topografiske kart i et lag-spesifikk mote 6. Om 38 typer medulla nevroner er til stede i hver medulla kolonne og forseggjorte dendritter i bestemte lag og med passende felt størrelser å motta innspill fra disse afferente 7. De synaptiske kretser i medulla har blitt rekonstruert ved elektronmikroskopi nivå; således er de synaptiske partnerskap vel etablert 7, 8. Videre genetiske verktøy for merking av forskjellige typer av medulla nevroner er tilgjengelige 9, 10, 11. Ved å undersøke tre typer transmedulla (TM) neuroner (Tm2, TM9 og TM20), har vi tidligere har identifisert to celletypespesifikke dendritiske egenskaper: (i) Tm neuroner rager dendritter i enten den fremre eller bakre retning (plan projEL retning), avhengig av celletyper og (ii) dendritter av medulla nevroner ender i bestemte medulla lag i en celletype-spesifikk måte (layer-spesifikk terminering) 12. Planar projeksjon retning og lag-spesifikke terminering er tilstrekkelig til å skille disse tre typer Tm nevroner, mens mutasjoner som forstyrrer Tm svar til lag og kolonne signaler påvirke ulike aspekter av disse attributtene. Her presenterer vi en komplett arbeidsflyt for å undersøke den dendrittiske fordelingen av Drosophila medulla nevroner i kolonner og lag (figur 1). Først viser vi en dual-view avbildningsmetode som bruker tilpasset programvare for å kombinere to confocal bilde stabler å generere høy kvalitet isotrope bilder. Denne metoden krever bare vanlig konfokalmikroskopi å generere bilder av høy kvalitet som gir mulighet for pålitelig sporing av dendrittiske grener, uten å ty til super-oppløsning mikroskopi, sliks STED (stimulert utslipp Nedbryting) eller strukturelle belysning. For det andre, presenterer vi en metode for å spore dendrittiske lysthus og for å registrere de resulterende neurite spor til en referanse kolonne matrise. For det tredje viser vi de beregningsmessige metoder for å ekstrahere informasjon om den plane projeksjon retning og lag-spesifikk opphør av dendritter, samt for å utlede estimater for dendritiske forgreninger og terminerings frekvenser. Sammen utgjør disse metodene gir mulighet for karakterisering av dendrittiske mønstre i 3D, klassifisering av celletyper basert på dendrittiske morfologi, og identifisering av mulige defekter i mutanter.

Arbeidsflyten omfatter tre deler: (i) dual-view bildeseksjonen kombinerer to confocal bildestakker samlet på ortogonale retninger i en høyoppløselig 3D-bilde av dendritter; (Ii) dendrite sporing og registrering § spor dendrittiskearbors i 3D og registrerer dendrittiske spor til referansekolonnen matrise; (Iii) det dendritiske analyse delen analyserer dendrittiske mønstre i forhold til kolonner og lag, inkludert laget spesifikk terminering og plane projeksjon retning av dendrittiske arbors, og utleder estimater av dendritiske forgreninger og terminerings frekvenser. Protokollene bruke tilpassede plugins bygget på åpen kildekode MIPAV (Medical Imaging Processing, analyse og visualisering) plattform og tilpassede verktøykasser i matrisen laboratoriet språk. Sammen utgjør disse protokollene tilveiebringe en komplett arbeidsflyt for å analysere den dendrittiske ruting av Drosophila medulla nevroner i lag og kolonner, for å identifisere celletyper, og for å bestemme defekter i mutanter.

Bruk av dual-view bildebehandling prosedyren som presenteres her, ble en flue hjernen inneholder tynt merket TM20 nevroner avbildes i to ortogonale retninger.
