Gist is een krachtige modelorganisme bij veroudering onderzoek. Echter, een gebruikelijke levensduur assay gist voert microdissectie, die niet alleen arbeidsintensief maar ook lage doorzet 1, 2. Bovendien is de traditionele microdissectie aanpak voorziet niet in een gedetailleerde weergave van verschillende cellulaire en moleculaire kenmerken van de alleenstaande moeder cellen als ze ouder worden. De ontwikkeling van microfluïdische apparaten is een geautomatiseerd systeem voor gist levensduur te meten en om moleculaire merkers en verscheidene cellulaire fenotypes volgen gedurende de levensduur van de moedercellen 3, 4, 5, 6, 7, 8 ingeschakeld. Na gistcellen in een microfluïdische inrichting zijn geladen, kunnen zij worden gevolgd onder een microscoop met automatische tijdsafhankelijkee beeldvorming. Met behulp van beeldvorming bewerkgereedschappen kunnen verschillende cellulaire en moleculaire fenotypes worden geëxtraheerd 3, 6, 8, zoals levensduur, formaat, fluorescente reporter, celmorfologie, cel cyclus dynamica, etc., waarvan vele zijn moeilijk of onmogelijk te verkrijgen met behulp de traditionele microdissectie methode. Microfluïdische apparaten hebben opgedaan bekendheid in gist veroudering onderzoek sinds hun succesvolle ontwikkeling van een paar jaar geleden 3, 4, 6, 7. Verschillende groepen zijn vervolgens gepubliceerd op variaties van de eerdere ontwerpen 5, en veel gist labs hebben microfluïdische apparaten gebruikt voor hun studie. In een celcultuur exponentiële groei ondergaan, het aantal bejaarde moeder cellen die voor observatie beschikbaar zijn, is miniscule. Daarom is het algemene ontwerpprincipe van de microfluïdische inrichting voor levensduur metingen te moedercellen behouden en dochtercellen te verwijderen. Een dergelijk ontwerp maakt gebruik van het feit dat gist ondergaat asymmetrische celdeling. De structuren in de inrichting wil houden groter moedercellen en laten kleinere dochtercellen weg te wassen. De microfluïdische chip in dit artikel wordt een zachte polydimethylsiloxaan (PDMS) pad (verticaal hangend kolommen) te controleren moedercellen (figuur 1). using microfluidic devices to measure lifespan and cellular phenotypes in single budding yeast cells

Budding yeast Saccharomyces cerevisiae is an important model organism in aging research. Genetic studies have revealed many genes with conserved effects on the lifespan across species. However, the molecular causes of aging and death remain elusive. To gain a systematic understanding of the molecular mechanisms underlying yeast aging, we need high-throughput methods to measure lifespan and to quantify various cellular and molecular phenotypes in single cells. Previously, we developed microfluidic devices to track budding yeast mother cells throughout their lifespan while flushing away newborn daughter cells. This article presents a method for preparing microfluidic chips and for setting up microfluidic experiments. Multiple channels can be used to simultaneously track cells under different conditions or from different yeast strains. A typical setup can track hundreds of cells per channel and allow for high-resolution microscope imaging throughout the lifespan of the cells. Our method also allows detailed characterization of the lifespan, molecular markers, cell morphology, and the cell cycle dynamics of single cells. In addition, our microfluidic device is able to trap a significant amount of fresh mother cells that can be identified by downstream image analysis, making it possible to measure the lifespan with higher accuracy.
