Colorimetrische cytotoxiciteitsbepalingen, zoals de 3- (4, 5-dimethyl-2-thiazolyl) -2,5-difenyl-2-H-tetrazoliumbromide (MTT) -assay, worden gewoonlijk gebruikt om de levensvatbaarheid van cellen in vitro te meten. Primaire gekweekte Cerebellar Granule Neuronen (CGN&#39;s) van ratten zijn gevoelig voor verschillende neurotoxinen, waaronder 1-methyl-4-fenylpyridinium-ion, waterstofperoxide en glutamaat 1 , 2 . Daarom kunnen CGN culturen gebruikt worden als een in vitro model op het gebied van neurowetenschappen. CGN-culturen kunnen een verscheidenheid aan cellen bevatten, waaronder neuronen en gliale cellen, die ongeveer 1% van de totale cellen in de CGN-cultuur kunnen uitmaken. Glialcellen reageren echter verschillend op neurotoxinen in vergelijking met neuronen, wat leidt tot een vooroordeel in de neuronale levensvatbaarheid gemeten door colorimetrische analyses 3 . In levensvatbare cellen kan fluoresceendiacetaat (FDA) omgezet worden in fluoresceïne door esterase.Propidiumjodide (PI) kan interageren met het DNA na het binnendringen van dode cellen en kan gebruikt worden om apoptose binnen de cultuur aan te geven. Daarom kan FDA-PI dubbele kleuring tegelijkertijd levensvatbare cellen en dode cellen evalueren, wat suggereert dat de cellevendigheid nauwkeuriger kan worden gemeten door beide colorimetrische methoden te combineren. Bovendien, door Hoechst toe te voegen, een blauwe fluorescerende vlek voor kernen, zou de nauwkeurigheid van de cellevendigheid verder kunnen worden verbeterd. Het hier beschreven protocol beschrijft FDA-PI dubbele kleuring en FDA-PI-Hoechst triple staining, die gebruikt kan worden om de neuronale levensvatbaarheid nauwkeurig te analyseren in primaire gekweekte CGN&#39;s. Dit protocol profiteert van het visualiseren en onderscheiden van CGNs en glialcellen door hun verschillende maten en vormen. Na kleuring worden de aantallen levensvatbare neuronen en dode neuronen geteld uit representatieve beelden die worden genomen door fluorescerende microscopie. De grote gliale cellen worden uitgesloten door de vergelijking van typische CGNs tAken onder fluorescerende modus met die welke onder fase contrast modus zijn genomen. Een soortgelijke strategie kan worden uitgevoerd om neuronale levensvatbaarheid in gemengde celculturen te meten die neuronen en gliale cellen bevatten, zoals primaire corticale culturen en hippocampale culturen.

Primaire gekweekte Cerebellar Granule Neuronen (CGN&#39;s) zijn veel gebruikt als een in vitro model in neuroscience en neuropharmacology onderzoek. Het co-bestaan ​​van gliale cellen en neuronen in CGN-cultuur kan echter leiden tot vooroordelen bij de nauwkeurige beoordeling van neuronale levensvatbaarheid. Fluoresceendiacetaat (FDA) en Propidiumjodide (PI) dubbele kleuring is gebruikt om de levensvatbaarheid van cellen te meten door de levensvatbare en dode cellen tegelijkertijd te evalueren. We hebben FDA-PI dubbele kleuring gebruikt om de gevoeligheden van de colorimetrische analyses te verbeteren en neuronale levensvatbaarheid in CGN&#39;s te evalueren. Dit protocol beschrijft hoe de nauwkeurigheid van de beoordeling van neuronale levensvatbaarheid kan verbeteren door gebruik te maken van deze methoden in de CGN-cultuur. Met behulp van dit protocol kan het aantal gliale cellen uitgesloten worden met behulp van fluorescentiemicroscopie. Een soortgelijke strategie kan worden toegepast om de ongewenste gli te onderscheidenAlcellen van neuronen in verschillende gemengde celculturen, zoals primaire corticale cultuur en hippocampale cultuur.

De dubbele immunostaining van GAP43 (rood) en GFAP (groen) werd gebruikt om de vorm van neuronen en gliale cellen, respectievelijk 2 , 5 te analyseren.
