Er zijn verschillende voorbeelden van biologische en biocompatibele synthetische materialen bestemd voor gebruik in celstudies en weefselregeneratie gericht op celhechting en proliferatie 1, 2, 3, 4, 5. Er zijn enkele voorbeelden van biocompatibele materialen, bekend als vloeibare kristallen elastomeren (LCEs), die kunnen reageren op externe stimuli anisotrope moleculaire bestelling 6, 7 zijn. LCEs zijn stimuli-responsieve materialen die de mechanische en elastische eigenschappen van elastomeren combineren met de optische functionaliteit en moleculaire ordening vloeibaar kristal 8, 9. LCEs kunnen vormveranderingen, mechanische vervorming, elastisch gedrag en optische eigenschappen ervaren in reactie op externe stimuli (bijv., warmte, stress, licht, enz.) 10, 11, 12, 13, 14, 15, 16. Eerdere studies hebben aangetoond dat vloeibare kristallen (LC) de groei en oriëntatie van cellen 4, 17 kan voelen. Men kan dan veronderstellen dat LCEs geschikt zijn voor biologisch en medisch relevante toepassingen, zoals mobiele steigers en uitlijning zijn. synthesis of biocompatible liquid crystal elastomer foams as cell scaffolds for 3d spatial cell cultures

Hier presenteren we een stap-voor-stap de bereiding van een 3D, biologisch afbreekbaar, schuim-achtige cel schavot. Deze skeletten werden bereid door verknoping sterblok copolymeren met eenheden cholesterol zijketen zijgroepen waardoor smectische-A (SMA) met vloeibare kristallen elastomeren (LCEs). Foam-achtige steigers, bereid met behulp van metalen sjablonen, voorzien van onderling verbonden microkanalen, waardoor ze geschikt zijn als 3D-celkweek steigers. De gecombineerde eigenschappen van de vaste structuur van het metaalschuim en het elastomeer resulteren in een 3D cel scaffold dat niet alleen een hogere celproliferatie bevordert vergeleken met gebruikelijke poreuze matrijs films, maar ook een beter beheer van massatransport (dwz voedingsstoffen, gassen, afval etc.). De aard van de metalen matrijs zorgt voor eenvoudige manipulatie schuim vormen (dat wil zeggen, rollen of films) en voor de bereiding van steigers met verschillende poriegrootten voor verschillende celstudies behoud van de interconnectieted poreuze aard van de matrijs. Het etsproces tast de chemie van de elastomeren, behoud van hun biologisch verenigbaar en biologisch afbreekbaar. We tonen aan dat deze smectische LCEs, wanneer ze groeien voor een uitgebreide tijdsperioden, schakelt u de studie van klinisch relevante en complexe weefsel constructies terwijl het bevorderen van de groei en proliferatie van cellen.

Dit rapport toont de bereidingswerkwijze van een poreuze 3D LCE als matrix voor celkweek onder toepassing van een nikkelmetaal template. De verkregen 3D LCE toont een complex onderling kanalennetwerk voor probleemloos celinfiltratie, alsmede geschikter massatransport 27.
