Hoewel structurele biologie werkwijzen, zoals röntgen kristallografie en NMR zijn voordelig omdat zij zeer gedetailleerd structuren van eiwitten, deze beelden vaak statisch. De karakterisering van voorbijgaande soorten en zwak gestructureerde domeinen blijft ongrijpbaar wanneer onderzocht door deze conventionele werkwijzen. In de afgelopen jaren zijn aanzienlijke vooruitgangen geboekt bij de ontwikkeling van analytische technieken ter eiwitstructuur en dynamica 1, 2, 3, 4 meten. Terwijl röntgenkristallografie blijft het principe voor het bepalen van eiwitstructuur worden hoge concentraties van eiwitten nodig en uitgebreide optimalisering vereist diffractie Kristallen produceren. Eiwitten die moeilijk te kristalliseren zijn, zoals membraangeassocieerd en intrinsiek ontvouwen eiwitten zijn klassiek bestudeerd door waterstof-deuterium uitwisseling (HDX) NMR 5. Echter, de laatste decennia koppeling van electrospray ionisatie massaspectrometrie (ESI-MS) naar HDX is snel aan populariteit 6, 7. Vooral MS zeer gevoelig (nM tot pM concentraties vereist), en er vrijwel geen grens aan eiwitgrootte. De ontwikkeling van tijdsopgeloste elektrosprayionisatie (Tresi) door Wilson en KONERMANN in 2003 toegestaan ​​reacties te kunnen houden in pseudo-real time door ESI-MS. time-resolved electrospray ionization hydrogen-deuterium exchange mass spectrometry for studying protein structure and dynamics

Intrinsiek ontvouwen eiwitten (IDP) zijn lang een uitdaging structurele biologen vanwege hun gebrek aan stabiele secundaire structuurelementen. Waterstof deuterium uitwisseling (HDX) gemeten bij snelle tijdschalen is bij uitstek geschikt voor structuren en waterstofbinding netwerken die kort worden bevolkt detecteren, waardoor de karakterisering van voorbijgaande conformeren in natieve ensembles. Koppeling van HDX aan massaspectrometrie biedt een aantal belangrijke voordelen, waaronder een hoge gevoeligheid, lage monster consumptie en geen beperking op eiwit grootte. Deze techniek is sterk gevorderd in de afgelopen tientallen jaren, inclusief de mogelijkheid om HDX etikettering de gaten te houden op de milliseconde tijdschaal.
