De hier beschreven protocol werd oorspronkelijk ontwikkeld om de locaties van twee subsets van CDC in de long zichtbaar. Echter, kan het protocol gemakkelijk worden aangepast aan vele verschillende celtypen te onderzoeken, met behoud van cellevensvatbaarheid en de driedimensionale architectuur van de long. De laatste is een belangrijk voordeel ten opzichte celkweeksystemen en vergemakkelijkt de identificatie van zeldzame celtypen. Na inhalatie van pro-inflammatoire stimuli zoals lipopolysaccharide (LPS), is er een gecoördineerde beweging van immuuncellen in, binnen en uit de longen. Bijvoorbeeld worden neutrofielen snel aangeworven om het longparenchym en luchtwegen. Bovendien hebben sommige professionele antigeenpresenterende cellen bekend als conventionele dendritische cellen (CDCS) ondergaat een betrekkelijk ingewikkelde migratiepatroon 1, 2. CDCS kunnen worden geïdentificeerd met gebruik van doorstroomcytometrie, gedeeltelijk gebaseerd op de weergave van de oppervlaktemerker, CD11c. Verschillende subsets van DCs kan worden onderscheiden door het verschil oppervlakte-expressie van CD103 en CD11b 3. Bij het verwerven van geïnhaleerde antigeen, sommige CDCs verlaat de long en migreren door de lymfevaten to-long drainerende lymfeklieren (LN), waar ze presenteren peptiden op antigeen-specifieke T-cellen 4. Dit is een kritieke vroege gebeurtenis bij de initiatie van adaptieve immuunsysteem responses. Om onbekende redenen, echter niet alle CDCs dat geïnhaleerde antigenen verwerven verlaat de longen, en veel van deze cellen blijven in dat orgaan voor enkele maanden 5, 6. Deze waarneming kan gedeeltelijk worden verklaard door de ontwikkelings afstamming van deze cellen omdat monocyten afgeleide CD11c + cellen die de chemokine receptor CCR7, niet kunnen migreren naar regionale LN&#39;s 7, 8. Het lijkt waarschijnlijk dat de migratiepotentieel van CDC Ook wordt bepaald, ten minste gedeeltelijk, door de anatomische positie binnen de long. Echter, de precieze lokalisatie van deze verschillende populaties van CDC in de long niet volledig gekarakteriseerd. Een verbeterde kennis van immuuncel lokalisatie binnen de longen en van de moleculen die direct het, is nodig voor een beter begrip van hoe het immuunsysteem van de longen wordt geactiveerd. PCLS worden dat verhogingly gebruikt als een ex vivo benadering voor cellulaire positionering en cel-cel interacties visualiseren, met behoud van de structurele integriteit van de long architectuur 9, 10. PCLS zijn gebruikt om de longen van de vele soorten, waaronder muizen, runderen, apen, schapen, paarden en mensen 11 te bestuderen. Een belangrijk voordeel van deze techniek is dat ongeveer 20 plakken kunnen worden bereid uit een kwab van een muislong, waardoor het aantal dieren om eigen proeven verminderen. Vrijwel alle immuun celtypen, met inbegrip van DCs, macrofagen, neutrofielen, en T-cellen, zijn aanwezig in PCLS en hun normale structuur te behouden. PCLS kan ook worden gebruikt om calcium signalering en contractiliteit van de luchtwegen en gladde spiercellen studie na behandeling met acetylcholine 12 of 13 methacholine. In deze benadering slechts een klein deel van de long is eenmicroscopisch alyzed, maar een studie rapporteerde dat metingen van de luchtwegen inkrimping PCLS varieert slechts ongeveer 10% van plak tot plak, en dit verschil is vergelijkbaar met dat gebruikt longfunctietesten bij intacte dieren 14. Andere onderzoekers hebben PCLS een ex vivo benadering gebruikt om veranderingen in cytokine-expressie en celoppervlak markers na incubatie studie met 15 LPS. PCLS zijn ook gebruikt in een ex vivo model van hypoxische pulmonale vasoconstrictie in kleine intra-acinaire slagaders. Deze vaten liggen in het deel van de long die niet kunnen worden bereikt met andere procedures, waaronder opnamen van ontleed arteriële segmenten of analyse van subpleural vaten 16. Ons laboratorium heeft voornamelijk gebruikt PCLS om immuuncel lokalisatie in levende longweefsel bij steady state en naar aanleiding van een in vivo inflammatoire stimulus te visualiseren. precision-cut mouse lung slices to visualize live pulmonary dendritic cells

Inhalatie van allergenen en pathogenen opwekt meerdere wijzigingen in diverse immunologische celsoorten in de longen. Flowcytometrie is een krachtige techniek voor kwantitatieve analyse van celoppervlakte-eiwitten op immuuncellen, maar geeft geen informatie over de lokalisatie en migratie patronen van deze cellen in de longen. Evenzo kan chemotaxis assays worden uitgevoerd om het vermogen van cellen te bestuderen om te reageren op chemotactische factoren in vitro, maar deze testen niet reproduceren complexe omgeving van het intacte long. In tegenstelling tot deze bovengenoemde technieken kan de ligging van de verschillende celtypes in de longen gemakkelijk worden gevisualiseerd door het genereren precisie gesneden Lung Schijfjes (PCLS) kleuring hen commercieel verkrijgbare, fluorescent gemerkte antilichamen en visualiseren van de secties met confocale microscopie. PCLS kan worden gebruikt voor zowel live als longweefsel gefixeerd en de plakjes kan gebieden omvatten zo groot als een dwarsdoorsnede van een hele kwab. We hebben dit protocol gebruikt met succes visualiseren van de locatie van diverse celtypen in de long, waaronder verschillende soorten dendritische cellen, macrofagen, neutrofielen, T-cellen en B-cellen, en structurele cellen zoals lymfatische, endotheliale en epitheelcellen. Het vermogen om cellulaire interacties, zoals die tussen dendritische cellen en T-cellen, in levende, driedimensionale longweefsel zichtbaar, kan openbaren hoe cellen zich binnen de longen met elkaar bij steady state en bij inflammatie. Derhalve bij gebruik in combinatie met andere procedures, zoals stroomcytometrie en kwantitatieve PCR, kan PCLS bijdragen aan een beter begrip van cellulaire gebeurtenissen die allergische en inflammatoire longziekten grondslag liggen.

Om de locatie van beide DC subsets, CD11b hi CDCs en CD103 + CDCs, PCLS van C57BL / 6-muizen te identificeren werden gesneden en gekleurd met monoklonale antilichamen (mAbs) die specifiek zijn voor CD11c, CD88, CD103 en CD324 (E-cadherine).
