Husets muskeshuggare , Suncus murinus , tillhör ordningen Insectivora och familjen Soricidae. Detta lilla däggdjur distribueras i hela Sydostasien och bevarar hus och gräsbevuxna områden som ligger nära mänskliga livsmiljöer eller nötkreaturpennor 4 . Denna art uppvisar allmänna morfologiska egenskaper som liknar människor än andra laboratoriedjur, såsom mus, råtta och kanin 5 . Tidigare användes helmonterad immunförstärkning med en perifer neuronmarkör för S. murinus neurofilamentprotein (NFP) för att märka perifera nerver i bukspottkörteln 6 , huvudduodenal papillan 7 , pylorus 8 , gallblåsan 5 och extrahepatisk gallvägar 9 . NFP är ett makromolekylärt komplex som ingår i den mogna neuronala cytoskeletten. Neurofilamentrelaterat proteinS mediera interaktioner mellan NFP och zygosomen. Både enzymfunktionen och strukturen hos linkerproteiner regleras av NFP 10 . NFP-komplexet är tillverkat av tre polypeptider: NF-L (70 kDa), NF-M (150-160 kDa) och NF-H (200 kDa). NFP kan hittas i neuronal axon i både det centrala och perifera nervsystemet. Anti-human NFP-antikropp har visat sig markera axoner i det centrala och perifera nervsystemet. Anatomiska och elektrofysiologiska undersökningar har visat att sfinkteren, gallblåsan och proximal mag-tarmkanalen är anslutna 11 , 12 , 13 , 14 . De morfologiska egenskaperna hos deras neuronala förbindelser har emellertid ännu inte definierats. Detta arbete demonstrerar användningen av en helmonterad immunohistokemisk färgmetod för att märka innervationen av S. murinus bilIarykan med en NFP-antikropp.

Detta arbete beskriver den fullständiga monteringen av immunhistokemiska färgmetoder i detalj, med användning av neurofilamentproteinantikroppar för att märka innervation av gallvägarna i Suncus murinus ( S. murinus &#160; ). Först dissekerades provet från S. murinus och fixerades i 4% paraformaldehyd (PFA). För det andra utfördes en enzymatisk behandling och potentiell endogen peroxidasinaktivering. Provet exponerades sedan för den primära antikroppen, anti-neurofilamentproteinantikroppen, i 3-6 dagar. Den inkuberades sedan med den sekundära antikroppen konjugerad med pepparrotsperoxidas. Färgreaktionen avslöts genom att reagera provet med ett 3,3&#39;-diaminobensidin (DAB) substrat. Denna metod kan tillämpas för att analysera innerveringen av alla viskosorgan. Vidare kan detta protokoll också anpassas för att testa andra neuronala antikroppar, men optimering av antibodiesEs bör göras först. Denna metod introducerades ursprungligen av Kuratani och Tanaka 1 , 2 , 3 .
