Channelrhodopsins (ChR) - световодные ионные каналы, которые происходят в пятне глаз подвижных зеленых водорослей, и служат в качестве первичных фотодатчиков для фототаксиса и фобических ответов 1 . С момента своего первого описания в 2002 году 2 , ChRs проложили путь к появляющейся области оптогенетики и могут применяться во множестве возбудимых клеток, например, в скелетных мышцах, сердце или головном мозге 3 , 4 , 5 . Применяемый в культуре клеток, этот метод обеспечивает точный контроль как ионных, так и электрических условий регистрации, а также позволяет исследовать селективность ионов наряду с относительным вкладом ионов в общий ток. Здесь мы проиллюстрируем исследование избирательности ионов для анион-проводящего канала rhodopsin Proteomonas sulcata ( Ps ACR1) 22 , 23 посредством регистрации вольт-амперных отношений при различных концентрациях внеклеточного хлорида, чтобы доказать высокую проводимость хлорида.

whole-cell patch-clamp recordings for electrophysiological determination of ion selectivity in channelrhodopsins

За последнее десятилетие каналодродоксины стали незаменимыми в неврологических исследованиях, где они используются как инструменты для неинвазивного манипулирования электрическими процессами в клетках-мишенях. В этом контексте особенно важна ионная селективность канала -родопсина. В данной статье описывается исследование селективности хлоридов для недавно идентифицированного анион-проводящего канала -родопсина Proteomonas sulcata посредством электрофизиологических записей патч-струбцины на клетках HEK293. Экспериментальная методика измерения светоизлучающих фототоков требует быстро переключаемого - идеально монохроматического - источника света, подключенного к микроскопу обычной установки патч-струбцины. Подготовительные процедуры перед экспериментом описаны с подготовкой буферных растворов, соображениями о потенциалах жидкого соединения, посевом и трансфекцией клеток и вытягиванием пипетки. Фактическая запись отношения напряжение-напряжениеС для определения обратимых потенциалов для различных концентраций хлоридов происходит через 24 ч до 48 ч после трансфекции. Наконец, электрофизиологические данные анализируются по теоретическим соображениям по хлоридной проводимости.

Whole-cell Patch-clamp Recordings for Electrophysiological Determination of Ion Selectivity in Channelrhodopsins

This article describes how the ion selectivity of channelrhodopsin is determined with electrophysiological whole-cell patch-clamp recordings using HEK293 cells. Here, the experimental procedure for investigating chloride selectivity of an anion-selective channelrhodopsin is demonstrated. However, the procedure is transferable to other channelrhodopsins of distinct selectivity.
