bioluminescence imaging of neuroinflammation in transgenic mice after peripheral inoculation of alpha-synuclein fibrils

Um das Prion-ähnliches Verhalten von falsch gefalteten alpha-Synuclein, Mausmodelle zu studieren benötigt werden, mit denen eine schnelle und einfache Übertragung von alpha-Synuclein prionoids, die Neuropathologie im Zentralnervensystem (ZNS) führen. Hier beschreiben wir das intraglossal oder intraperitoneale Injektion von alpha-Synuclein - Fibrillen in bigenic Tg (M83 +/-: GFAP -Luc +/-) Mäuse, die humanen alpha-Synuclein mit der A53T - Mutation aus dem Prionprotein - Promotor überexprimieren und Glühwürmchen - Luciferase aus der Promotor für gliales fibrilläres saures Protein (GFAP), ist ausreichend , um neuropathologische Erkrankung zu induzieren. Im Vergleich zu homozygot Tg (M83 + / +) Mäusen , die Anfang schwere neurologische Symptome im Alter von 8 Monaten, heterozygot Tg (M83 +/-: GFAP -Luc +/-) entwickeln Tiere bleiben frei von spontaner Krankheit , bis sie ein erreichen Alter von 22 Monaten. Interessanterweise ist die Injektion von alpha-Synuclein-Fibrillen über die intraperitoneMäuse , die mit einer medianen Inkubationszeit von 229 ± 17 Tagen: al Route neurologische Erkrankung mit Paralyse in vier von fünf Tg (GFAP -Luc +/- +/- M83) induziert. Erkrankte Tiere zeigten schwere Ablagerungen von phosphoryliertem alpha-Synuclein in ihrem Gehirn und Rückenmark. Anhäufungen von alpha-Synuclein waren Sarkosyl-unlösliche und mit Ubiquitin und p62 colokalisiert, und wurden durch eine Entzündungsreaktion begleitet, was zu astrozytären Gliose und Mikrogliose. Überraschenderweise Inokulation von alpha-Synuclein-Fibrillen in die Zunge war weniger wirksam bei der Entstehung von Krankheiten mit nur einer der fünf injizierten Tieren alpha-Synuclein Pathologie nach 285 Tagen zeigen. Unsere Ergebnisse zeigen , dass die Impfung über die intraglossal Strecke und um so mehr über die intraperitoneale Route geeignet ist neurologische Erkrankung mit entsprechenden Kennzeichen Synucleinopathien in Tg (M83 +/-: GFAP -Luc +/-) zu induzieren Mäuse. Dies stellt ein neues Modell für die Untersuchung Prion-ähnliche Pathogenese induziertD durch alpha-Synuclein prionoids näher.
