Hier tonen we een wijd toegankelijke methodologie om de rol van kinase signaalwegen te onderzoeken bij het reguleren van naïeve-primed pluripotente transitie. Dit behandelt een sleutelvraag in het ESC-veld. Hoewel de doorstromingsgenetica en transcriptomics benaderingen routinematig worden gebruikt om belangrijke transcriptie-regulatoren van naïeve en primitieve pluripotente staten te identificeren, blijkt uit te leggen dat pluripotentie-signaleringsnetwerken uitdagend zijn. We bieden nu een flexibele strategie aan die chemische remmers gebruikt om kinases te identificeren die de naïeve-primed pluripotente overgang in mESC&#39;s beheersen. We gebruiken eenvoudige mESC-cultuuromstandigheden en standaard reagentia, materialen en apparatuur om kinase-remmers te ontdekken en te valideren met tot nu toe onverminderd vermogen om naïeve pluripoten te stabiliseren. a simple method to identify kinases that regulate embryonic stem cell pluripotency by high-throughput inhibitor screening

Embryonale stamcellen (ESC&#39;s) kunnen zelf vernieuwen of differentiëren in alle celsoorten, een fenomeen dat bekend staat als pluripotentie. Bepaalde pluripotente staten zijn beschreven, genaamd &quot;naïeve&quot; en &quot;primed&quot; pluripotency. De mechanismen die de naïeve-gepresteerde overgang beheersen zijn slecht begrepen. In het bijzonder blijven we slecht geïnformeerd over proteïne kinasen die naïeve en priide pluripotente staten specificeren, ondanks de toenemende beschikbaarheid van hoogwaardige gereedschapverbindingen om de kinasefunctie te onderzoeken. Hier beschrijven we een schaalbaar platform om gerichte kleine molecuulschermen uit te voeren voor kinase-regulatoren van de naïeve-primed pluripotente transitie in muis-ESC&#39;s. Deze aanpak maakt gebruik van eenvoudige celcultuuromstandigheden en standaard reagentia, materialen en apparatuur om kinaseremmers te ontdekken en te valideren met tot nu toe niet-gemarkeerde effecten op pluripotentie. We bespreken potentiële applicaties voor deze technologie, waaronder het screenen van ander klein molecuulCollecties zoals steeds meer geavanceerde kinaseremmers en opkomende bibliotheken van epigenetische hulpmiddelenverbindingen.

Met behulp van de hier geplaatste procedure ( Figuur 1 ), screenen we de LINCS-bibliotheek van 228 krachtige en selectieve kinase-remmers om degenen te identificeren die mESC pluripotency moduleren. De bibliotheek wordt bereid in een concentratie van 0,1 mM voor een verdunning van 1: 100 en eindscreening van 1 uM in mESC&#39;s. 48 uur later werden mESC&#39;s gelyserd en extracten bereid voor kwantitatieve dot blot analyse van Nanog en Dnmt3b expressie ( Figuur 2 , boven). Resultaten van andere screeningsconcentraties worden hier niet voor kortheid weergegeven. Nanog: Dnmt3b verhouding wordt bepaald voor elke kinaseremmer en verbindingen gerangschikt en een 2x cut-off is toegepast ( Figuur 2 , bodem). Totaal Nanog- en Dnmt3b-signaal ten opzichte van de controle wordt overgebracht op de ranglijst van de inhibitor en onderworpen aan een 0,5x cut-off, om trimmering van remmers mogelijk te maken die significante mESC toxiciteit vertonen. Selecteer kinaseremmers die de naïeve sta stabiliserenTe worden gevalideerd met behulp van conventionele Nanog / Dnmt3b immunoblotting ( Figuur 3 ). De primaire kinase doelen van deze remmers worden weergegeven in tabel 1 . We tonen ook de toepasbaarheid van dit scherm aan om remmers te identificeren die de primed toestand 14 stabiliseren. <img alt="Figuur 1" src="/files/ftp_upload/55515/55515fig1.jpg" /> Figuur 1: Workflow voor het screenen van kinase remmers die naïeve-primed transitie moduleren.
