Hier presenteren we chimera assemblage door plasmide recovery en restriction enzyme site insertion (CAPRRESI). CAPRRESI profiteert van vele sterke punten van de oorspronkelijke plasmide herstel methode en introduceert restrictie enzym digestie om DNA ligatie reacties te vereenvoudigen (vereist voor chimera assemblage). Voor dit protocol, klonen gebruikers wildtype genen in hetzelfde plasmide (pUC18 of pUC19). Na de in silico selectie van aminozuursequentie gebieden waar chimera&#39;s moeten worden samengesteld, krijgen gebruikers alle synoniem-DNA-sequenties die ze coderen.
