Therapeutische antilichamen zijn recombinante monoklonale antilichamen (mAbs) ontwikkeld voor de behandeling van verschillende pathologieën, waaronder kanker, auto-immuun en chronische ziekten, neurologische stoornissen en anderen 1 . Momenteel heeft de FDA goedkeuring verleend aan meer dan 40 therapeutische mAbs, en er wordt verwacht dat de markt de komende jaren zal bereiken. Rituximab is een chimerisch monoklonaal IgG1-antilichaam met hoge affiniteit, goedgekeurd voor de behandeling van CD20 + B-cel niet-Hodgkin&#39;s lymfoom (NHL), CD20 + folliculaire NHL, chronische lymfocytische leukemie en reumatoïde artritis 2 , 3 . Bijgevolg moeten de methoden die in dergelijke vergelijkingen worden gebruikt, de structurele en functionele eigenschappen van de mAb&#39;s beoordelen, met name die met klinische relevantie. Daartoe tonen in vitro analyses verschillende voordelen ten opzichte van in vivo experimenten (beoordeeld in Chapman et al. ) 5 : i) in vitro studies zijn gevoeliger voor verschillen tussen de voorgestelde biosimilar en het referentieproduct; Ii) in vivo studies moeten worden uitgevoerd in relevante soorten, die voor veel mAbs zijnniet-menselijke primaten; en iii) omdat het werkingsmechanisme, preklinische toxicologie en klinische effecten van het referentieproduct zijn welbekend in vivo studies met biosimilaire kunnen aanvullende nuttige informatie wordt gegeven. Therapeutische monoklonale antilichamen (mAbs) zijn op de behandeling van verschillende ziekten, waaronder kanker betrokken. De ontwikkeling van de biosimilar mAbs door farmaceutische bedrijven is een markt kans, maar het is ook een strategie om de drug de toegankelijkheid te verhogen en-therapie bijbehorende kosten te verlagen. De protocollen hier beschreven beschrijven de evaluatie van target binding en CDC inductie door rituximab in Daudi cellen. Dit protocol beschrijft de in vitro vergelijking van twee belangrijke functionele kenmerken van rituximab: target binding en complement-dependent cytotoxicity (CDC) inductie. De methoden werden gebruikt voor een vergelijking tussen referentie rituximab en een rituximab biosimilar. Deze analyses kunnen worden gebruikt bij biosimilar ontwikkeling of als kwaliteitscontrole in hun productie.
