Er zijn verscheidene alternatieve werkwijzen voor het inbrengen kleine RNA&#39;s in zoogdiercellen, die in het algemeen in chemische, biologische of fysische categorieën 10 vallen. Gewoonlijk gebruikte chemische werkwijzen, waaronder op lipiden gebaseerde transfecties en calciumfosfaat transfectie, afhankelijk maken van chemisch-DNA-complexen die efficiënter door cellen genomen. Over het algemeen chemische werkwijzen niet zo efficiënt voor de transfectie van primaire T-cellen. De meest voorkomende biologische methode is om een virale vector (bijvoorbeeld retrovirus of lentivirus), dat direct ingevoegd vreemde RNA in een gastheer als onderdeel van zijn natuurlijke replicatiecyclus gebruiken. Virale transductie duurt kenmerkend langer in beslag, vooral wanneer men factoren in de tijd voor moleculaire klonering van provirale plasmiden. Daarnaast kunnen virale vectoren transductie potentieel schadelijk voor de onderzoekers. Elektroporatie is een fysiek methode induceren membraan permeabilisatie door het onderwerpen van cellen aan hoogspanningspulsen, waarbij nucleïnezuren transiënt voeren in de cel waar ze inwerken op hun doel. Traditionele elektroporatie instrumenten waren niet effectief voor het transfecteren primaire lymfocyten. Toch is geoptimaliseerd next generation elektroporatie bewezen in staat T-cellen te transfecteren met zeer hoge efficiëntie, in het bijzonder wanneer het materiaal te transfecteren klein RNA. De term generatie losjes gebruikt om de twee nieuwere platforms (bijvoorbeeld neon, Amaxa) door traditionele elektroporatie machines differentiëren. Bovendien is deze werkwijze gemakkelijk schaalbaar voor matige doorvoer screenings met tot ongeveer 120 kleine RNAs in een enkel experiment, vaak met gevalideerde synthetische reagentia. Belangrijker is dat succesvolle transfecties worden bereikt in slechts 16 uur na T-celactivering. Het nadeel van deze methode is echter dat zij niet tot stabiele genomische incorporatiop, en is daarom van voorbijgaande aard. Daarom is de extra inspanning een stabiel expressieconstruct dat kan worden verpakt in een virale vector en met succes tot expressie gebracht in T-cellen wanneer langdurige expressie van een kleine RNA nodig maken waard. We hebben een nieuwe generatie transfectie (bijvoorbeeld Neon) gebruikt om diverse synthetische enkel- of dubbelstrengs RNA of LNA oligonucleotide gereedschappen leveren voor verschillende doeleinden 11, 12, 13. Efficiënte RNA interferentie kan worden geïnduceerd in primaire muis en menselijke T-cellen met behulp van dubbelstrengs korte interfererende RNA (siRNA). Dit protocol beschrijft geoptimaliseerde omstandigheden voor het gebruik van deze techniek in humane cellen Th17. In aanvulling op siRNA, kunnen in de handel verkrijgbare synthetische miRNA bootst en remmers worden gebruikt om miRNA winst en verlies van functie te bestuderen. miRNA bootst dubbelstrengs RNA-moleculen, vergelijkbaar met siRNA, maar designed met de sequentie van endogeen mature miRNAs. miRNA-remmers worden chemisch gemodificeerde RNA en / of LNA gebaseerd enkelstrengs oligonucleotiden die binden aan natief miRNAs en hun functie antagoniseren. CD4+ T cells can differentiate into several subsets of effector T helper cells depending on the surrounding cytokine milieu. Th17 cells can be generated from na&#239;ve CD4+ T cells in vitro by activating them in the presence of the polarizing cytokines IL-1&#946;, IL-6, IL-23, and TGF&#946;. Th17 cells orchestrate immunity against extracellular bacteria and fungi, but their aberrant activity has also been associated with several autoimmune and inflammatory diseases. Th17 cells are identified by the chemokine receptor CCR6 and defined by their master transcription factor, ROR&#947;t, and characteristic effector cytokine, IL-17A. Optimized culture conditions for Th17 cell differentiation facilitate mechanistic studies of human T cell biology in a controlled environment. They also provide a setting for studying the importance of specific genes and gene expression programs through RNA interference or the introduction of microRNA (miRNA) mimics or inhibitors. This protocol provides an easy to use, reproducible, and highly efficient method for transient transfection of differentiating primary human Th17 cells with small RNAs using a next generation electroporation device.
