Talrige rapporter har vist, at calprotectin har antimikrobielle aktiviteter mod forskellige patogener, herunder Staphylococcus aureus , Candida albicans , Acinetobacter baumannii , Klebsiella pneumoniae , Escherichia coli og H. pylori , og at de inhiberende virkninger skyldes metalcheleringsaktiviteten af ​​calprotectin 25 , 28 , 29 . H. pylori kræver zink, som det blev bestemt ved tidligere forskning, der anvendte et kemisk defineret medium for at fastslå mikronæringsstofkravene for dette patogen for at vokse og proliferere 34 . Derudover var calprotectin meget rigeligt inden for H. pylori- inficerede væv og associeret med neutrofile infiltrater, hvilket indikerer at værten kan anvende calprotectin som en antimikrobiell strategi under infektion og efterfølgende inflammation 25 , 35 . Nylige bevis fra objektive proteomics screeningsteknikker antyder, at under betingelser, hvor reaktive iltarter er rigelige, calprotecTin gennemgår posttranslationelle modifikationer, som ændrer hexa-histidinbindingsstedet og derved hæmmer proteinbindelsens metalbindende aktivitet. Som sådan antydede vi, at andre S100A-familieproteiner blandt de brede repertoar af disse molekyler kunne potentielt fungere som hjælpemetalkelatorer. Vi valgte S100A12 til videre undersøgelse, fordi den ikke blev identificeret i den ovennævnte skærm for posttranslationel modifikation, den har kapacitet til at binde zink, og den er meget rigelig i humane væv stammer fra H. pylori- inficerede individer. Vores arbejde indikerer, at S100A12 kan hæmme H. pylori- vækst og levedygtighed på en dosisafhængig måde i H. pylori- G27-stammen, og at den antimikrobielle aktivitet af dette protein kan reverseres ved tilsætning af overskydende næringsstofsink. Dette arbejde supplerer vores tidligere arbejde med angivelse af S100A12 udøvet antimikrobiel aktivitet mod PMSS1 og 7.13-stammer af H. pylori , der demonstrerer sin brede antibakterielle aktivitet mod talrige kliniske isolater og laboratorie-tilpassede stammer af H. pylori 18 . Sammen bekræfter disse resultater betydningen af ​​S100A12 som en mekanisme til at kontrollere bakteriel vækst og proliferation via ernæringsimmunitet. Fremtidige undersøgelser af denne vigtige værtsbakterieinteraktion kan omfatte udnyttelse af aktiviteten af ​​S100A12 for at reducere bakteriel byrde i værtsvæv eller bestemmelse af dette proteins bidrag til immun signalering i forbindelse med H. pylori- infektion.
