S100 eiwitten zijn een klasse van de EF-hand familie van calcium bindende eiwitten met een uiteenlopende reeks functies 1 . Ze worden uitgedrukt in een weefsel- en celspecifieke manier en regelen een breed spectrum van cellulaire functies 2 , 3 . Uniek voor calciumbindende eiwitten, S100-eiwitten vertoonden zowel intracellulaire als extracellulaire functies 4 , 5 . Binnen de cel veroorzaakt Ca 2+ binding een conformatieve verandering die een hydrofoob oppervlak blootstelt dat specifiek op eiwitbindende partners 6 staat . Dit intracellulaire mechanisme regelt belangrijke processen zoals cel proliferatie, differentiatie en energie metabolisme. In het extracellulaire milieu hebben S100 eiwitten twee functies 7 . In een, fungeren ze als schade geassocieerde moleculaire patroon (DAMP) eiwitten en initiëren een pro-inflammatoireAtoom immuunrespons door interactie met patroonherkenningsreceptoren 8 , 9 . Daarnaast hebben meerdere leden van de S100 proteïne klasse sequester overgangsmetalen, een functie die dient om microbiële pathogenen te verhongeren in een proces genaamd voedingsimmuniteit 10 , 11 . S100A12 (ook bekend als calgranulin C en EN-RAGE) wordt sterk uitgedrukt in macrofagen en neutrofielen en is geïdentificeerd als een potentiële biomarker voor ontstekingsziekten 12 , 13 . Naast bindend calcium op zijn EF-Hand-plaatsen heeft S100A12 twee hoge affiniteits-overgangsmetaalbindingsplaatsen, gelegen aan tegenover elkaar gelegen uiteinden van de dimerinterface 14 , 15 . Calprotectine is een neutrofiele geassocieerde eiwit die een heterodimeer vormt van de S100A8- en S100A9-subeenheden die overgangsmetalen binden aan de dimerinterface 31 . Calprotectine is aangetoond om twee metaalbindingsplaatsen te bezitten: Site 1 kan Zn 2+ , Mn 2+ , of Fe 2+ binden en Site 2 kan<html> Bind Zn 2+ 31 , 32 . Verschillende rapporten hebben aangetoond dat calprotectin antimicrobiële activiteiten tegen diverse pathogenen heeft, waaronder Staphylococcus aureus , Candida albicans , Acinetobacter baumannii , Klebsiella pneumoniae , Escherichia coli en H. pylori , en dat de remmende effecten zijn te wijten aan de metaalchelatieactiviteit van calprotectin 25 , 28 , 29 . Eerdere werkzaamheden aangetoond dat calprotectine talrijke activiteiten op H. pylori uitoefent, inclusief het veranderen van de lipide A-structuur in het buitenmembraan, onderdrukking van het cag- Type IV-secretiesysteem (dat een belangrijke proinflammatoire virulentiefactor binnen H. pylori is ), het induceren van biofilmvorming en het onderdrukken van H. pylori groei en levensvatbaarheid op een dosisafhankelijke manierClass = &quot;xref&quot;&gt; 25 , 33 . Bovendien bleken genetische en biochemische analyses de antibacteriële activiteit van calprotectine tegen H. pylori te berusten, grotendeels afkomstig van zijn vermogen om nutriëntzink 25 te binden. H. pylori vereist zink, zoals werd bepaald door eerder onderzoek dat een chemisch gedefinieerd medium gebruikt om de micronutriëntvereisten voor dit pathogeen te bepalen om 34 te groeien en te prolifereren. Daarnaast was calprotectine zeer overvloedig in H. pylori- geïnfecteerde weefsels, en geassocieerd met neutrofiele infiltraten, wat aangeeft dat de gastheer calprotectine kan gebruiken als een antimicrobiële strategie tijdens infectie en daaropvolgende ontsteking 25 , 35 . Recent bewijs van onbevooroordeelde proteomische screeningstechnieken suggereert dat onder omstandigheden waarin reactieve zuurstofsoorten overvloedig zijn, calprotecTin ondergaat post-translationele modificaties die de hexa-histidine bindingsplaats veranderen, waardoor de metaalbindende activiteit van het eiwit 36 wordt geremd. Als zodanig hebben we verondersteld dat andere eiwitten van de S100A-familie onder het brede repertoire van deze moleculen potentieel kunnen optreden als hulpmetaal-chelators. We hebben S100A12 geselecteerd voor verdere studie, omdat het niet werd geïdentificeerd in het voornoemde scherm voor posttranslatorische modificatie, het heeft de capaciteit om zink te binden en het is zeer overvloedig in menselijke weefsels afgeleid van H. pylori- geïnfecteerde individuen. Ons werk geeft aan dat S100A12 de groei en de levensvatbaarheid van H. pylori op dosisafhankelijke wijze kan remmen in de G27-stam van H. pylori en dat de antimicrobiële activiteit van dit eiwit omgekeerd kan worden door toevoeging van overtollig nutriëntzink. Dit werk aanvult ons vorige werk dat S100A12 aangeeft dat antimicrobiële activiteit tegen PMSS1 en 7.1 heeft uitgeoefend3 stammen van H. pylori , die zijn brede antibacteriële activiteit aantonen tegen talrijke klinische isolaten en laboratorium aangepaste stammen van H. pylori 18 . Samen bevestigen deze resultaten het belang van S100A12 als een mechanisme om bacteriële groei en proliferatie via voedingsimmuniteit te beheersen. Toekomstige studies van deze belangrijke gastro-bacteriële interactie zouden onder meer de uitoefening van de activiteit van S100A12 kunnen omvatten om de bacteriële last in gastheerweefsels te verminderen of de bijdrage van dit eiwit aan immuun signalering in de context van H. pylori- infectie te bepalen.

Calgranulin eiwitten zijn belangrijke mediators van aangeboren immuniteit en zijn leden van de S100 klasse van de EF-hand familie van calcium bindende eiwitten. Sommige S100-eiwitten hebben de mogelijkheid om overgangsmetalen te binden met een hoge affiniteit en ze kunnen ze effectief wegvallen van invallende microbiële pathogenen in een proces dat &quot;voedingsimmuniteit&quot; wordt genoemd. S100A12 (EN-RAGE) bindt zowel zink als koper en is zeer overvloedig in aangeboren immuuncellen, zoals macrofagen en neutrofielen. We rapporteren een verfijnde methode voor de expressie, verrijking en zuivering van S100A12 in zijn actieve, metaalbindende configuratie. Gebruik van dit eiwit in bacteriële groei en levensvatbaarheidsanalyses laat zien dat S100A12 antimicrobiële activiteit heeft tegen het bacteriepatogen, Helicobacter pylori . De antimicrobiële activiteit is gebaseerd op de zinkbindende activiteit van S100A12, die chloorvoedingszink chelaat, waardoor H. pylori verhongert, wat zink vereist voor groei en proliferation.
