Bisherige Arbeiten zeigten, dass Calprotektin zahlreiche Aktivitäten auf H. pylori ausübt, einschließlich der Veränderung der Lipid-A-Struktur in der äußeren Membran und unterdrückt das Cag-Typ IV-Sekretionssystem (das ein wichtiger proinflammatorischer Virulenzfaktor innerhalb von H. pylori ist ), wodurch die Biofilmbildung und die Unterdrückung induziert wird H. pylori Wachstum und Lebensfähigkeit in einer dosisabhängigen WeiseClass = &quot;xref&quot;&gt; 25 , 33 . Darüber hinaus zeigten genetische und biochemische Assays die antibakterielle Aktivität von Calprotectin gegen H. pylori wurde weitgehend aus seiner Fähigkeit, Nährstoff Zink 25 zu binden abgeleitet. Der jüngste Beweis von den unvoreingenommenen Proteomik-Screening-Techniken deutet darauf hin, dass unter Bedingungen, bei denen reaktive Sauerstoffspezies reichlich vorhanden sind, calprotecZinn unterliegt posttranslationalen Modifikationen, die die Hexa-Histidin-Bindungsstelle verändern und dadurch die Metallbindungsaktivität des Proteins 36 hemmen. Als solches haben wir vermutet, dass andere S100A-Familienproteine ​​unter dem breiten Repertoire dieser Moleküle potentiell als Hilfsmetall-Chelatoren wirken könnten. Wir haben S100A12 für eine weitere Studie ausgewählt, weil es in dem oben erwähnten Screen für die posttranslationale Modifikation nicht identifiziert wurde, es hat die Fähigkeit, Zink zu binden, und es ist sehr reich an menschlichen Geweben, die von H. pylori- infizierten Individuen stammen. Unsere Arbeit zeigt, dass S100A12 H. pylori Wachstum und Lebensfähigkeit in einer dosisabhängigen Weise im G27-Stamm von H. pylori hemmen kann und dass die antimikrobielle Aktivität dieses Proteins durch die Zugabe von überschüssigem Nährstoff-Zink umgekehrt werden kann. Diese Arbeit ergänzt unsere bisherige Arbeit, die darauf hinweist, dass S100A12 antimikrobielle Aktivität gegen PMSS1 und 7.1 ausübt3-Stämme von H. pylori , die ihre breite antibakterielle Wirkung gegen zahlreiche klinische Isolate und Labor-adaptierte Stämme von H. pylori 18 zeigen . Gemeinsam bestätigen diese Ergebnisse die Bedeutung von S100A12 als Mechanismus zur Kontrolle des Bakterienwachstums und der Proliferation durch Ernährungsunempfindlichkeit. Zukünftige Untersuchungen dieser wichtigen Wirtsbakterien-Interaktion könnten die Ausnutzung der Aktivität von S100A12 zur Verringerung der Bakterienbelastung innerhalb von Wirtsgeweben oder die Bestimmung des Beitrags dieses Proteins zur Immunsignalisierung im Kontext der H. pylori- Infektion umfassen.
