Les récepteurs couplés aux protéines G (GPCR) sont une grande famille de récepteurs de surface cellulaire responsables d&#39;une gamme remarquable de processus physiologiques, y compris l&#39;analgésie, l&#39;olfaction et le comportement 1 . Les GPCR agissent en détectant des signaux externes spécifiques et en stimulant ensuite la signalisation intracellulaire. Ils marquent donc une jonction clé entre les environnements externe et interne d&#39;une cellule. En raison du rôle critique que jouent les GPCR dans la biologie, ils sont devenus des cibles majeures pour la recherche fondamentale et la découverte de médicaments 2 , 3 . Contrairement à d&#39;autres familles de récepteurs qui se lient à des ligands discrets, les GPCR peuvent lier des molécules très différentes. Alors qu&#39;un GPCR peut interagir avec des peptides, un autre peut détecter les photons, les petites molécules ou les ions 1 , 4 . Bien que leurs ligands soient diversifiés, les GPCR sont unifiés dans leur architecte généralUre et fonction. Les GPCR individuels sont constitués de sept protéines transmembranaires α-hélicoïdales avec des terminaux amino extracellulaires et des terminaux carboxyle intracellulaires 5 , 6 . Les GPCR sont couplés aux protéines G intracellulaires - complexes de protéines hétérotrimères composés de sous-unités α, β et γ - qui opèrent sur différentes voies de signalisation 7 . La sous-unité G α est une protéine de liaison aux nucléotides de guanine qui est inactif lorsqu&#39;elle est liée au diphosphate de guanosine (PIB) et active lorsqu&#39;elle est liée au guanosine triphosphate (GTP) 8 , 9 . Lorsque les GPCR lient leurs ligands, ils subissent un changement conformationnel qui permet à G α de se dissocier de G βγ , permettant ainsi à G α d&#39;échanger le PIB pour GTP 7 . Le récepteur lui-même est phosphorylé à son extrémité carboxyle par divers serine / threonIne kinases 10 , 11 et internalisées pour atténuer la signalisation du récepteur 12 , 13 , 14 . Pendant ce temps, le monomère G α activé et le dimère G βγ procèdent à l&#39;activation de voies de signalisation distinctes 7 . Il existe plusieurs isoformes de chaque sous-unité de protéine G, et chaque isoforme cible des voies en aval et des systèmes de messagerie secondaire particuliers. Les principales isoformes de G α comprennent G s , G q , G i / o et G 12-13 . Généralement, les GPCR individuels s&#39;associaient à une isoforme G α particulière, reliant ainsi un stimulus externe à une réponse cellulaire spécifique 1 . La caractérisation d&#39;une interaction GPCR-ligand est essentielle pour comprendre la biologie du récepteur. Comme l&#39;échange de PIB / GTP est l&#39;une des premières veilleNts qui suit la liaison du ligand, la surveillance de la liaison GTP peut mesurer l&#39;activation ou l&#39;inhibition du GPCR. L&#39;analyse de plus d&#39;événements en aval dans la signalisation GPCR n&#39;est souvent pas aussi quantitatif ni stoechiométrique, peut ne pas distinguer les agonistes complets de ceux partiels et peut nécessiter des réactifs coûteux. En outre, l&#39;augmentation de la liaison GTP aux protéines G α est un événement presque universel suite à l&#39;activation du GPCR, ce qui signifie que la mesure de la liaison GTP est un dosage largement applicable pour le suivi de l&#39;activité de la plupart des GPCR. La mesure de la liaison GTP est une approche simple et rapide pour surveiller la signalisation GPCR dans les cellules surexprimant le récepteur d&#39;intérêt ou dans le tissu indigène. Le présent protocole détaille un dosage fonctionnel de liaison GTP en utilisant un GPCR archétypal, le récepteur μ-opioïde (MOR1), pour déterminer quantitativement l&#39;activité d&#39;un agoniste et d&#39;un antagoniste sur la signalisation GPCR. Ce protocole décrit d&#39;abord comment isoler les membranes brutes des cellules qui surexpriment MOR1. Notez queCe protocole ne se limite pas aux systèmes de surexpression et peut être appliqué à de nombreuses sources de membrane, y compris des tissus indigènes ou des préparations exprimant des récepteurs multiples et des protéines G 15 . Le protocole détaille ensuite comment mesurer la liaison d&#39;un analogue GTP radioactif à ces membranes en réponse à des concentrations variables de [D-Ala, N-MePhe, Gly-ol] -enkephalin (DAMGO) ou de naloxone, un agoniste et antagoniste MOR1, respectivement. L&#39;analogue GTP, [ 35 S] guanosine-5&#39;-O- (3-thio) triphosphate ([ 35 S] GTPγS) n&#39;est pas hydrolysable. Cette propriété est critique parce que les sous-unités G α présentent une activité GTPase intrinsèque 7 et élimineraient le phosphate gamma marqué sur un radiochimique GTP hydrolysable. Les membranes sont ensuite piégées sur des filtres à fibres de verre et lavées, après quoi le GTP radiomarqué est quantifié par comptage par scintillation liquide. Des paramètres pharmacologiques multiples peuvent être dérivés pour caractériserE l&#39;interaction récepteur-ligand, y compris la réponse demi-maximale (EC 50 ) et le coefficient Hill (n H ) pour les agonistes et la concentration inhibitrice demi-maximale (IC 50 ) et la constante de dissociation à l&#39;équilibre (K b ) pour les antagonistes 16 , 17 , 18 .

Les récepteurs couplés à la protéine G (GPCR) sont une grande famille de récepteurs transmembranaires qui jouent un rôle critique dans la physiologie cellulaire normale et constituent une cible pharmacologique majeure pour de multiples indications, y compris l&#39;analgésie, la régulation de la pression artérielle et le traitement des maladies psychiatriques. Lors de la liaison au ligand, les GPCR catalysent l&#39;activation des protéines G intracellulaires en stimulant l&#39;incorporation du guanosine triphosphate (GTP). Les protéines G activées stimulent alors les voies de signalisation qui provoquent des réponses cellulaires. La signalisation GPCR peut être surveillée en mesurant l&#39;incorporation d&#39;une forme radiomarquée et non hydrolysable de GTP, [ 35 S] guanosine-5&#39;-O- (3-thio) triphosphate ([ 35 S] GTPγS), en protéines G. Contrairement à d&#39;autres méthodes qui évaluent plus de processus de signalisation en aval, [ 35 S] GTPγS binding mesure un événement proximal dans la signalisation GPCR et, surtout, peut distinguer l&#39;agonisTs, antagonistes et agonistes inverse. Le présent protocole décrit une méthode sensible et spécifique pour l&#39;étude de la signalisation GPCR en utilisant des préparations à membrane brute d&#39;un GPCR archétypal, le récepteur μ-opioïde (MOR1). Bien qu&#39;il existe des approches alternatives pour le fractionnement des cellules et des tissus, beaucoup sont coûteuses, fastidieuses et / ou nécessitent des équipements de laboratoire non standard. La présente méthode fournit une procédure simple qui enrichit les membranes brutes fonctionnelles. Après avoir isolé MOR1, diverses propriétés pharmacologiques de son agoniste, [D-Ala, N-MePhe, Gly-ol] -enkephalin (DAMGO) et antagoniste, naloxone, ont été déterminées.
