I recettori G-Proteine-Coupled (GPCRs) sono una grande famiglia di recettori della superficie cellulare responsabile di una notevole serie di processi fisiologici, tra cui analgesia, olfazione e comportamento 1 . I GPCR agiscono rilevando segnali esterni specifici e successivamente stimolando la segnalazione intracellulare. Esse pertanto rappresentano una giunzione chiave tra gli ambienti esterni e interni di una cella. A causa del ruolo critico che i GPCR svolgono in biologia, sono diventati obiettivi principali sia per la ricerca di base che per la scoperta di droga 2 , 3 . A differenza di altre famiglie di recettori che legano ligandi discreti, i GPCRs possono legare tipi molto diversi di molecole. Mentre un GPCR può interagire con i peptidi, un altro può sentire i fotoni, le piccole molecole oi ioni 1 , 4 . Mentre i loro ligandi sono diversi, GPCRs sono unificati nel loro architetto globaleUre e funzione. La subunità G α è una proteina legante nucleotidica guanina che è inattiva quando legata al guanosina difosfato (PIL) e attiva quando legata al guanosina trifosfato (GTP) 8 , 9 . I recettori G-Proteine-Coupled (GPCRs) sono una grande famiglia di recettori transmembranti che svolgono un ruolo critico nella normale fisiologia cellulare e costituiscono un importante obiettivo farmacologico per molteplici indicazioni, tra cui l&#39;analgesia, la regolazione della pressione arteriosa e il trattamento delle malattie psichiatriche. Su legame vincolante, i GPCR catalizzano l&#39;attivazione delle proteine ​​G intracellulari, stimolando l&#39;incorporazione di guanosina trifosfato (GTP). Le proteine ​​G attivate stimolano quindi i percorsi di segnalazione che provocano risposte cellulari. La segnalazione di GPCR può essere monitorata misurando la incorporazione di GTP, [35S] guanosina-5&#39;-O- (3-tio) trifosfato ([ 35 S] GTPγS) in proteine ​​G, di una forma radiolabelata e non idrolizzabile. Il presente protocollo descrive un metodo sensibile e specifico per studiare la segnalazione GPCR utilizzando preparazioni a membrana grezze di un GPCR archetipo, il recettore μ-opioide (MOR1). Anche se esistono approcci alternativi a frazionamento di cellule e tessuti, molti sono cost-prohibitive, noiosi e / o richiedono apparecchiature di laboratorio non standard.
