G- 단백질 결합 수용체 ( G-Protein-Coupled Receptor , GPCRs)는 정상적인 세포 생리학에서 중요한 역할을하는 막 투석 수용체의 큰 계열이며 진통, 혈압 조절 및 정신병 치료와 같은 여러 적응증의 주요 약리학 적 표적을 구성합니다. 리간드 결합시, GPCR은 구아 노신 트리 포스페이트 (GTP)의 혼입을 자극하여 세포 내 G- 단백질의 활성화를 촉매한다. 활성화 된 G- 단백질은 세포 반응을 유도하는 신호 전달 경로를 자극합니다. GPCR 시그널링은 G- 단백질로 [ 35 S] 구아노 신 -5&#39;-O- (3- 티오) 트리 포스페이트 ([ 35 S] GTPγS)의 방사성 표지 및 비 가수 분해성 형태의 결합을 측정함으로써 모니터 할 수있다. 더 많은 하류 신호 전달 과정을 평가하는 다른 방법과 달리, [ 35 S] GTPγS binding은 GPCR 신호 전달에서 근위 사건을 측정하며, 중요하게는,ts, 길항제 및 역작용 제를 포함한다. 현재의 프로토콜은 원형 GPCR, μ-opioid 수용체 (MOR1)의 조질 막 준비를 사용하여 GPCR 신호를 연구하기위한 민감하고 구체적인 방법을 개략적으로 설명합니다. 세포와 조직을 분별하는 대안적인 방법이 있지만, 비용이 많이 들고, 지루하고, 비표준 실험 장비가 필요합니다. 본 방법은 기능성 조 (crude) 막을 풍부하게하는 간단한 절차를 제공한다.
