Autologe chondrocyt implantatie (ACI) is een relatief nieuwe, state-of-the-art procedure voor het behandelen van articulair kraakbeen defecten 1 , 2 . Een van de cruciale stappen in ACI is de amplificatie van chondrocyten via de monolagcultuur in vitro . Tijdens amplificatie verlielen de hyaline chondrocyten hun fenotype gemakkelijk en worden gedifferentieerd, wat ongewenst is voor ACI-behandeling 3 , 4 . Om de uitkomst van ACI-behandeling te optimaliseren, moet de mate van chondrocytedifferentiatie worden bepaald voor replantatie. Het is essentieel om een ​​economische en snelle manier vast te stellen om de status van chondrocyten te bepalen. Onlangs heeft de associatie tussen DNA methylatie en chondrocyt dedifferentiatie veel aandacht aangetrokken 4 , 5 , 6 . DNA methylering is een proces van whDe methylgroepen worden toegevoegd aan DNA, wat resulteert in de omzetting van cytosine residuen naar 5-methylcytosine (5-mC). Om de biologie van DNA-methylering bij chondrocytedifferentiatie te verduidelijken, is de eerste stap het DNA-methyleringsniveau van chondrocyten, dat tot nu toe uitdagend is, te evalueren. Bisulfiet genomische sequencing is de meest gebruikte techniek om DNA-methylering 7 , 8 te analyseren. Bij deze analyse veroorzaakt bisulfietomzetting DNA-afbraak en moet dus een substantiële hoeveelheid monster voor de bepaling worden verschaft. Ook is een high-performance vloeistofchromatografie (HPLC) gebruikt om de globale DNA-methyleringsniveaus 9 , 10 te kwantificeren. HPLC analyse vereist echter genomische DNA-vertering. Bovendien zijn geavanceerde en dure experimentele instrumenten nodig. Daarom zijn deze experimentele procedures naast de hoge kosten tijdsverschillenuming. Anti-5-mC-antilichamen zijn nu in de handel verkrijgbaar, wat de mogelijkheid tot immuun blotting van 5-mC-bevattend genoom DNA van complexe genen heeft gecreëerd. In dit rapport hebben we genomisch DNA uit chondrocyten geëxtraheerd in een serie monolagenculturen geëxtraheerd. We gebruikten een dot blot analyse om het 5-mC gehalte in humane chondrocyten met verschillende aantallen passages te evalueren. We hebben geconstateerd dat 5-mC-inhoud is verhoogd in sterk gedifferentieerde chondrocyten in vergelijking met chondrocyten met laagwaardige dedifferentiatie.
