L&#39;implantation de chondrocytes autologue (ACI) est une procédure relativement nouvelle et à la fine pointe de la technique pour traiter les défauts du cartilage articulaire 1 , 2 . L&#39;une des étapes cruciales de l&#39;ACI est l&#39;amplification des chondrocytes par culture monocouche in vitro . Pendant l&#39;amplification, les chondrocytes hyalines perdent facilement leur phénotype et deviennent dédifférenciés, ce qui n&#39;est pas souhaitable pour le traitement ACI 3 , 4 . Pour optimiser le résultat du traitement ACI, l&#39;étendue de la dédifférenciation des chondrocytes devrait être déterminée avant la replantation. Il est impératif d&#39;établir un moyen économique et rapide de déterminer le statut des chondrocytes. Récemment, l&#39;association entre la méthylation de l&#39;ADN et la dédifférenciation des chondrocytes a attiré beaucoup d&#39;attention 4 , 5 , 6 . La méthylation de l&#39;ADN est un processus par whDes groupes méthyle sont ajoutés à l&#39;ADN, ce qui entraîne la conversion des résidus de cytosine en 5-méthylcytosine (5-mC). Pour élucider la biologie de la méthylation de l&#39;ADN dans la dédifférenciation des chondrocytes, la première étape consiste à évaluer le niveau de méthylation de l&#39;ADN des chondrocytes qui, jusqu&#39;à présent, se sont révélés difficiles. Le séquençage génomique du bisulfite est la technique la plus utilisée pour analyser la méthylation de l&#39;ADN 7 , 8 . Dans ce dosage, la conversion de bisulfite provoque une dégradation de l&#39;ADN, et donc une quantité substantielle d&#39;échantillon doit être fournie pour l&#39;essai. En outre, la chromatographie liquide haute performance (HPLC) a été utilisée pour quantifier les niveaux globaux de méthylation de l&#39;ADN 9 , 10 . Cependant, l&#39;analyse HPLC nécessite une digestion par l&#39;ADN génomique. En outre, des instruments expérimentaux avancés et coûteux sont nécessaires. Par conséquent, en plus du coût élevé, ces procédures expérimentales sont des contre-inconvénientsUming. Les anticorps anti-5-mC sont maintenant disponibles dans le commerce, ce qui a créé la possibilité d&#39;un transfert immunologique d&#39;ADN génomique contenant 5 mC à partir de génomes complexes. Dans ce rapport, nous avons extrait l&#39;ADN génomique des chondrocytes cultivés dans une série de cultures monocouches. Nous avons utilisé un test de point blot pour évaluer le contenu de 5 mC dans les chondrocytes humains avec différents nombres de passages. Nous avons constaté que le contenu de 5 mC était augmenté dans les chondrocytes fortement dédifférenciés par rapport aux chondrocytes à dédifférenciation de bas grade. En outre, nous avons identifié une relation entre le statut de dédifférenciation et les niveaux de 5 mC. Enfin, nous avons signalé que les changements dans le contenu de 5 mC étaient associés au phénotype des chondrocytes. Par conséquent, l&#39;essai de transfert de point de 5 mC est une méthode fiable, simple et rapide pour détecter le niveau de méthylation de l&#39;ADN dans les chondrocytes.

Le niveau global de méthylation de l&#39;ADN des chondrocytes est considéré comme un biomarqueur approprié pour la perte du phénotype des chondrocytes. Cependant, les résultats basés sur différentes méthodes expérimentales peuvent être incohérents. Par conséquent, il est important d&#39;établir une méthode précise, simple et rapide pour quantifier les niveaux globaux de méthylation de l&#39;ADN pendant la dédifférenciation des chondrocytes. Les techniques actuelles d&#39;analyse de la méthylation à l&#39;échelle du génome reposent largement sur le séquençage génomique des bisulfites. En raison de la dégradation de l&#39;ADN pendant la conversion du bisulfite, ces procédés nécessitent généralement un volume d&#39;échantillon important. Dans cette approche dot blot, un mélange d&#39;ADNg contenant l&#39;ADN méthylé à détecter a été repéré directement sur une membrane N + en tant que point à l&#39;intérieur d&#39;un modèle de modèle circulaire préalablement étiré. Par rapport à d&#39;autres approches d&#39;imprégnation à base d&#39;électrophorèse sur gel et d&#39;autres procédures complexes de transfert, la méthode de transfert de points enregistre un temps important. En outre, les transferts de points peuvent détecter le niveau global de méthylation de l&#39;ADN en utilisant un anticorps 5-mC disponible dans le commerce.
