Hvordan fornemmer organismer og svare skadelige temperaturer er stadig dårligt forstået. Endvidere mekanismerne bag sensibilisering af den sensoriske maskiner, såsom hos patienter med perifer neuropati eller skadeinducerede sensibilisering, er ikke godt karakteriseret. Den genetisk medgørlig Drosophila model er blevet anvendt til at undersøge cellerne og gener, der kræves for skadelige varme påvisning, hvilket har givet flere bevarede gener af interesse. Der vides imidlertid om cellerne og receptorer er vigtige for skadelige kold sensing. Selv Drosophila ikke overlever langvarig udsættelse for kolde temperaturer (≤10 ºC), og vil undgå køligt, men foretrækker varmere temperaturer i adfærdsmæssige præference assays, hvordan de fornemmer og muligvis undgå skadelige kolde stimuli er først for nylig blevet undersøgt. Her beskriver og karakterisere den første skadelige kolde (≤10 ºC) adfærdsmæssige assay viDrosophila. Ved hjælp af dette værktøj og analyse, viser vi en efterforsker, hvordan man kvalitativt og kvantitativt vurdere kolde nociceptive adfærd. Dette kan gøres under normale / sunde dyrkningsbetingelser, eller antagelig i forbindelse med sygdom, skade eller sensibilisering. Endvidere kan dette assay anvendes på larver valgt for ønskede genotyper, som kunne tænkes at påvirke thermosensation, smerte eller nociceptiv sensibilisering. Eftersom smerte er et særdeles konserveret proces, ved hjælp af dette assay til yderligere undersøgelse termisk nociception vil sandsynligvis indsamle vigtig forståelse af smerte processer i andre arter, herunder hvirveldyr.

Her demonstrerer vi en hidtil ukendt assay til at studere kold nociception i Drosophila larver.
