Drosophila har vist seg å være svært nyttig for identifisering av nye konserverte gener og neuronal kretser som ligger til grunn komplekse problemer. Fluer tilveiebringe en sofistikert genetisk verktøykasse og et forenklet nervesystemet som gir mulighet for nøyaktig genetisk manipulasjon og neuronal 1, 2, 3, 4 for å dissekere de cellulære og molekylære baser av nociception 5, 6, 7. Analysen beskrevet her gjør det mulig for den quantification av enten normal eller potensielt forandres atferdsmessige responser for lave temperaturer som strekker seg fra skadelige kald (≤ 10 ° C), ufarlige kule (11-17 ° C), til omgivelsestemperatur (18-22 ° C). De lave temperaturene som anvendes i denne analysen er i stand til direkte å aktivere klasse III sensoriske neuroner, utløsning av robuste, reproduserbare kalsium øker og kald-fremkalte atferdsmessige responser, som kan være kvalitativt og kvantitativt analysert 7. Gitt at thermosensation er allestedsnærværende i mange arter, gir denne analysen et verdifullt verktøy for studier av nocicepsjon og kan avdekke nye genmål eller neuronal interaksjoner som vil forbedrevår forståelse av virveldyr nociception. Den spesialbygde kald probe (se kald probe, Table of Materials) benytter en lukket sløyfe temperaturkontrollert Peltier-enhet, som kjøler aluminium akselen og den koniske spiss gjennom varmeledning. Hvordan organismer oppfatte og svare på skadelige temperaturer er fortsatt dårlig forstått. Videre er mekanismene bak sensibilisering av sensoriske maskineri, slik som hos pasienter med perifer nevropati eller skader indusert sensitivering, er ikke godt karakterisert. Den genetisk medgjørlig Drosophila modell er blitt brukt til å studere celler og gener som er nødvendige for skadelige varmedetektor, som har gitt multiple konserverte gener av interesse.
