このプロトコルの重要なステップは、エマルジョンの均質性を保証することです。よく調製されたエマルジョンでは、抗原は完全に油でコーティングされ、注入された抗原の徐放および持続的な免疫刺激を確実にする。このプロトコルのもう1つの重要な特徴は、ルイスラットの使用である。ルイスラットは、他の株27よりも自己免疫疾患の発症に対して感受性が高い。 このプロトコルは、涙腺抽出物のみまたは組換えKlk1b22 20,21のいずれかを使用した、以前に公開された2つのプロトコルから変更されている。現在のプロトコールでは、オボアルブミン+涙腺抽出物が抗原として使用され、抗原特異的免疫細胞が眼球表面および涙腺に引き寄せられ、局所組織損傷を誘発する。ドライアイはゆっくりと発達し、最初の免疫感作の48日後に約85％に達する。抗原への挑戦48日目に眼および涙腺が乾いた目を悪化させ、70日目まで慢性を確実にする。 Klk誘発性DEDモデルにおける組換えKlk1b22と比較して、現在のモデルで使用される涙腺抽出物およびオボアルブミンは、安価で入手しやすい。涙腺抽出物には、Klk以外にも自己免疫を誘導する可能性のある他のタンパク質が含まれているため、この抽出物はDED誘発時のKlk法よりも理論的に強力です。我々はまた、涙腺抽出物のみをラットに免疫した。これらの免疫したラットはDEDを発症したが、対照と比較して眼球組織におけるエフェクター記憶T細胞の有意な増加はなかった。 この手法の限界は、モデルを達成するのに70日かかることです。エフェクター記憶T細胞は、正常なラットの眼における主要なT細胞サブセットである。このモデルでは、自己免疫DEDは、眼球におけるCD3 +エフェクターメモリーT細胞の増加をもたらす。あの薬したがって、Kv1.3カリウムチャネルの選択的阻害剤などのエフェクター記憶T細胞を効果的に抑制することは、自己免疫DED28に対する治療上の利益を有し得る。

ドライアイ疾患（DED）は、涙および眼球表面の多因子性疾患であり、不快感、視覚障害および涙液膜不安定の症状をもたらし、眼表面の損傷を引き起こす可能性がある。これは、涙液膜の浸透圧の増加および眼表面1の炎症を伴う。 DEDに伴う症状は、燃焼、刺すような痛み、異物感、涙、眼の疲労、および乾燥である 2,3 。 DEDの2つの主な原因は、涙分泌腺による涙液産生の減少および涙液膜の過剰な蒸発である4 。シェーグレン症候群、全身性エリテマトーデス、およびリウマチ様関節炎のような自己免疫疾患を有する患者において、マイボーム腺に対する免疫損傷は、涙の安定に必須の脂質の発現を減少させる。また、眼球表面への免疫損傷は、表面濡れ性のために重要なムチンの存在。一緒に、これらのプロセスは累積的に慢性的なドライアイを引き起こす5,6,7 。 涙の補充および抗炎症療法は治療の主役です。しかしながら、DED（ すなわち、コルチコステロイドおよびシクロスポリン）の現在の抗炎症療法は、広く免疫抑制性であり、深刻な副作用をもたらす8,9,10 。自己免疫ドライアイを治療するための新規な免疫調節剤を試験するのに適した動物モデルが必要である。 特定の遺伝子欠損を有するマウス11,12,13 、特定の遺伝子を欠くマウス14,15、および免疫調節を過剰発現するトランスジェニックマウスory遺伝子は、自己免疫ドライアイのモデルとして用いられている16,17 。抗原誘発自己免疫動物モデルも、マウス18 、ウサギ19 、およびラット20,21 において報告されている。ここでは、慢性自己免疫ドライアイの抗原誘発モデルを記載する。このモデルは、以前の2つのモデルの変更です。 1つは涙腺抽出物を使用し、第2のものは涙腺20,21から自己抗原（ すなわち klk1b22）を使用した。 この疾患は、オボアルブミン、完全フロイントアジュバント、およびSprague-Dawleyラットの涙腺抽出物を含むエマルジョンを含む6〜8週齢のメスのLewisラット（ 図1 ）の皮下免疫によって誘導された。不完全フロイントアジュバント中の同じ抗原による2回目の免疫化は、2週間後に投与した。抗原特異的免疫細胞を涙腺および眼表面に補充するために、涙腺抽出物およびオボアルブミン（1mg / mL）の混合物を、6〜7週目に結膜下結膜および涙腺に注入した（ 図1 ）。ラットの85％以上が初回免疫の70日後にドライアイの特徴的な特徴を発達させた。これらの特徴には、涙液産生の減少（ 図2 ）、角膜フルオレセイン染色の増加（ 図3 ）、および涙液の安定性低下（ 図4 ）が含まれる。フローサイトメトリーによる正常ラットの眼球におけるT細胞の免疫プロファイリングは、CD3 +エフェクターメモリーT細胞の優勢を明らかにする（ 図5および6 ）。自己免疫DEDを有するラットは、CD3 +エフェクターメモリーT細胞の増加およびナイーブおよび中央メモリーT細胞の対応する減少を示す（ 図6 </strong&gt;）。

ドライアイ疾患は、罹患率および健康管理の負担を引き起こし、生活の質を低下させる非常に一般的な状態である。自己免疫ドライアイ状態を治療するための新規な治療法を試験するためには、適切なドライアイ動物モデルが必要である。このプロトコルは、慢性自己免疫ドライアイラットモデルを記述する。ルイスラットを、涙腺抽出物、オボアルブミンおよび完全フロイントアジュバントを含むエマルジョンで免疫化した。不完全フロイントアジュバント中の同じ抗原による2回目の免疫化を2週間後に行った。これらの免疫感作は、尾の基部に皮下投与した。眼表面および涙腺における免疫応答を高めるために、涙腺抽出物およびオボアルブミンを、初回免疫の6週間後に結膜下結膜および涙腺に注入した。ラットは、涙液の生成の減少、涙の安定性の低下、および角膜の損傷の増加を含むドライアイの特徴を発達させた。免疫プロフローサイトメトリーによるファイリングは、眼球におけるCD3 +エフェクター記憶T細胞の優勢を示した。
