Chemotaxis, ein Prozess der gerichteten Zellmigration zu löslichem chemischen Konzentrationsgradienten, ist kritisch in viele biologische Prozesse involviert, einschließlich Immunantwort 1 , 2 , 3 , Gewebeentwicklung 4 und Krebsmetastase 5 . Neutrophile sind die häufigsten weißen Blutzellen-Untergruppe und spielen entscheidende Rollen in der Ermöglichung der körpereigenen Gastgeber Verteidigung Funktionen, sowie bei der Vermittlung der adaptiven Immunantwort 6 , 7 . Neutrophile sind mit hochregulierten chemotaktischen Maschinen ausgestattet, die es ermöglichen, dass diese beweglichen Immunzellen sowohl auf Pathogen-abgeleitete Chemoattraktanten ( zB fMLP) als auch auf Wirts-abgeleitete Chemoattraktanten ( zB Interleukin-8) durch die Chemotherapie 8 reagieren. Neutrophile Migration und Chemotaxis vermitteln verschiedene physiologische ProblemeUnd Krankheiten wie Entzündungen und Krebs 1 , 9 . So liefert die genaue Beurteilung der neutrophilen Chemotaxis eine wichtige funktionelle Auslesung für das Studium der Neutrophilenbiologie und der damit verbundenen Erkrankungen. Im Vergleich zu den weit verbreiteten konventionellen Chemotaxis-Assays ( zB Transwell-Assay 10 ) zeigen die mikrofluidischen Geräte ein großes Versprechen für die quantitative Auswertung von Zellmigration und Chemotaxis aufgrund der präzise kontrollierten chemischen Gradientenbildung und Miniaturisierung 11 , 12 , 13 . In den letzten zwei Jahrzehnten wurden verschiedene mikrofluidische Geräte entwickelt, um die Chemotaxis verschiedener biologischer Zelltypen, insbesondere der Neutrophilen, zu untersuchen. Es wurde erhebliche Anstrengungen zur Charakterisierung der Neutrophilen-Migration in räumlich-zeitlich komplexem Che gegeben<html> Mischen Gradienten, die in den mikrofluidischen Vorrichtungen 14 , 15 konfiguriert wurden. Interessante Strategien wurden auch entwickelt, um eine direktionale Entscheidungsfindung durch Neutrophile unter Verwendung der mikrofluidischen Vorrichtungen zu untersuchen. An der biologisch orientierten Forschung wurden die Anwendungen von mikrofluidischen Vorrichtungen erweitert, um klinische Proben für die Krankheitsauswertung 17 , 18 , 19 zu testen. Allerdings ist die Verwendung von vielen mikrofluidischen Geräten auf spezialisierte Forschungslaboratorien beschränkt und erfordert eine lange neutrophile Isolierung aus großen Mengen an Blutproben. Daher hat es einen wachsenden Trend der Entwicklung integrierter mikrofluidischer Vorrichtungen zur schnellen Neutrophilen-Chemotaxis-Analyse direkt aus einem Tropfen Vollblut 20 , 21 , 22 ,Ef &quot;&gt; 23 , 24 . In dieser Richtung wurde ein All-on-Chip-Verfahren entwickelt, das die magnetische Negativ-Neutrophilen-Reinigung und den darauffolgenden Chemotaxis-Assay auf einer einzigen mikrofluidischen Vorrichtung 25 integriert . Diese All-on-Chip-Methode hat die folgenden neuartigen Merkmale: 1) Im Gegensatz zu früheren On-Chip-Strategien, die Neutrophile aus dem Blut durch adhäsionsbasierte Zellaufnahme oder zellgrßenbasierte Filterung 20 , 22 isolieren, erlaubt diese neue Methode hohe Reinheit, On-Chip-Magnettrennung der Neutrophilen aus kleinen Volumina von Vollblut sowie Chemotaxis-Messung bei chemoattraktiver Stimulation; 2) Die Zell-Docking-Struktur hilft, die Anfangspositionen der Neutrophilen in der Nähe des chemischen Gradientenkanals auszurichten und ermöglicht eine einfache Chemotaxisanalyse ohne Einzelzellverfolgung; 3) die Integration der Neutrophilen-Isolation und des ChemotsAchsen-Assay auf einer einzigen mikrofluidischen Vorrichtung ermöglicht eine schnelle Proben-zu-Ergebnis-Chemotaxis-Analyse in 25 min, wenn es keine Unterbrechung zwischen experimentellen Schritten gibt. Dieses Papier bietet ein detailliertes Protokoll für die Konstruktion, Betrieb und Datenanalyse Methode dieser All-on-Chip-Chemotaxis-Assay. Das Papier zeigt die effektive Verwendung des entwickelten Verfahrens zur Durchführung von Neutrophilen-Chemotaxis durch Testen eines bekannten rekombinanten Chemoattraktans und komplexer chemotaktischer Proben von Patienten, gefolgt von einer Diskussion über Strategien zur Fehlerbehebung, Einschränkungen und zukünftigen Richtungen.

an all-on-chip method for rapid neutrophil chemotaxis analysis directly from a drop of blood

Neutrophile Migration und Chemotaxis sind entscheidend für das Immunsystem unseres Körpers. Mikrofluidische Geräte werden zunehmend zur Untersuchung von Neutrophilenmigration und Chemotaxis aufgrund ihrer Vorteile in der Echtzeit-Visualisierung, der präzisen Kontrolle der chemischen Konzentrationsgradientenerzeugung und des reduzierten Reagenz- und Probenverbrauchs eingesetzt. In jüngster Zeit wurde von den mikrofluidischen Forschern zunehmend Anstrengungen unternommen, integrierte und leicht bedienbare mikrofluidische Chemotaxis-Analysesysteme direkt aus Vollblut zu entwickeln. In dieser Richtung wurde das erste All-on-Chip-Verfahren zur Integration der magnetischen Negativreinigung von Neutrophilen und des Chemotaxis-Assays aus kleinen Blutvolumenproben entwickelt. Diese neue Methode erlaubt einen schnellen Stichproben-Neutrophilen-Chemotaxis-Test in 25 min. In diesem Papier bieten wir detaillierte Bau-, Betriebs- und Datenanalyseverfahren für diesen All-on-Chip-Chemotaxis-Assay mit einer Diskussion über Strategieschutzstrategien, LimiUnd zukünftige Richtungen. Repräsentative Ergebnisse des Neutrophilen-Chemotaxis-Assays, die ein definiertes Chemoattraktant, N- Formyl-Met-Leu-Phe (fMLP) und Sputum von einem chronisch obstruktiven Lungenkrankheit (COPD) -Patienten unter Verwendung dieses All-on-Chip-Verfahrens untersuchen, werden gezeigt. Diese Methode gilt für viele zellmigrationsbezogene Untersuchungen und klinische Anwendungen.

Neutrophile werden aus einem Tropfen Vollblut direkt in der mikrofluidischen Vorrichtung negativ ausgewählt. Die Reinheit der isolierten Neutrophilen wurde durch On-Chip-Giemsa-Färbung verifiziert und die Ergebnisse zeigten die typischen ringförmigen und lobenförmigen Kerne von Neutrophilen ( Abbildung 2A ) 25 .
