Chemotaxis, en prosess med rettet cellemigrasjon til oppløselig kjemisk konsentrasjonsgradient, er kritisk involvert i mange biologiske prosesser, inkludert immunrespons 1 , 2 , 3 , vevsutvikling 4 og kreftmetastase 5 . Neutrofiler er den mest vanlige hvite blodlegemundersatsen og spiller viktige roller for å muliggjøre kroppens medfødte vertsforsvarsfunksjoner, samt å formidle adaptive immunresponser 6 , 7 . Neutrofiler er utstyrt med høyregulert kjemotaktisk maskineri slik at disse motile immunceller kan reagere på både patogen-avledede kjemoattraksjonanter ( f.eks. FMLP) og vert-avledede kjemoattraktanter ( f.eks . Interleukin-8) gjennomkjemotaks 8 . Neutrofile migrering og kjemotaksis medierer ulike fysiologiske problemerOg sykdommer som betennelse og kreft 1 , 9 . Dermed gir den nøyaktige vurderingen av nøytrofil kjemotaksis en viktig funksjonell avlesning for å studere nøytrofilbiologien og de tilknyttede sykdommene. Sammenlignet med de allment brukte konvensjonelle kjemotaksanalysene ( f.eks. Transwellanalyse 10 ), viser de mikrofluidiske innretninger godt løfte for kvantitativ evaluering av cellemigrasjon og kjemotaksis på grunn av den nøyaktig kontrollerte kjemiske gradientgenerering og miniaturisering 11 , 12 , 13 . I løpet av de siste to tiårene har ulike mikrofluidiske enheter blitt utviklet for å studere kjemotaksen av forskjellige biologiske celletyper, spesielt nøytrofile 11 . Interessante strategier ble også utviklet for å studere retningsbestemt avgjørelse ved nøytrofiler ved bruk av mikrofluidiske enheter 16. Ved siden av biologisk orientert forskning har applikasjonene av mikrofluidiske enheter blitt utvidet til testkliniske prøver for sykdomsevaluering 17 , 18 , 19 . Imidlertid er bruken av mange mikrofluidiske enheter begrenset til spesialiserte laboratorier og krever langvarig nøytrofilisolasjon fra store mengder blodprøver. Derfor har det vært en økende trend med å utvikle integrerte mikrofluidiske enheter for rask nøytrofilt kjemotaksanalyse direkte fra en dråpe fullblod 20 , 21 , 22 ,Ef &quot;&gt; 23 , 24 . Mot denne retningen ble det utviklet en all-on-chip-metode som integrerer den magnetiske negative nøytrofilrensningen og den påfølgende kjemotaksanalyse på en enkelt mikrofluidisk anordning 25 . Denne all-on-chip-metoden har følgende nye funksjoner: 1) I motsetning til tidligere on-chip-strategier som isolerer nøytrofiler fra blodet ved adhesjonsbasert cellefangst eller cellestørrelsesfiltrering 20 , 22 , tillater denne nye metoden høye Renhet, magnetisk separasjon på nøytropiler fra små volumer av helblod, samt kjemotaksemåling ved kjemoattraktantstimulering; 2) celledockingsstrukturen hjelper til med å justere startposisjonene til nøytrofilene nær den kjemiske gradientkanalen og tillater enkel kjemotaksanalyse uten enkeltcellesporing; 3) integrasjon av nøytrofilisolasjon og kjemotAkseanalysen på en enkelt mikrofluidisk enhet tillater rask analyse av kjemotaksanalyser i løpet av 25 minutter når det ikke er noen avbrudd mellom eksperimentelle trinn. an all-on-chip method for rapid neutrophil chemotaxis analysis directly from a drop of blood

Neutrofile migreringer og kjemotaksier er kritiske for kroppens immunsystem. Mikrofluidiske enheter brukes i økende grad for å undersøke neutrofilmigrasjon og kjemotaks, på grunn av deres fordeler i sanntidsvisualisering, presis kontroll av kjemisk konsentrasjonsgradientgenerering, og redusert reagens og prøveforbruk. Nylig er det gjort en økende innsats av de mikrofluidiske forskerne mot utvikling av integrerte og lettstyrte mikrofluidiske kjemotaksanalysesystemer, direkte fra helblod. I denne retning ble den første all-on-chip-metoden utviklet for å integrere den magnetiske negative rensingen av nøytrofiler og kjemotaksanalysen fra små blodvolumprøver. Denne nye metoden tillater en rask prøve-til-resultat nøytrofil kjemotaksis test på 25 minutter. I dette papiret gir vi detaljert konstruksjon, drift og dataanalysemetode for denne all-on-chip chemotaxis-analysen med en diskusjon om feilsøkingsstrategier, limiTasjoner og fremtidige retninger. Representative resultater av nøytrofilt kjemotaksis-analysen som tester en definert kjemoattraktant, N- formyl-Met-Leu-Phe (fMLP) og sputum fra en pasient med kronisk obstruktiv lungesykdom (COPD), ved bruk av denne all-on-chip-metoden, er vist. Denne metoden gjelder for mange celle-migrasjonsrelaterte undersøkelser og kliniske anvendelser.

Neutrofiler er negativt valgt fra en dråpe helblod direkte i den mikrofluidiske enheten. Renheten av de isolerte neutrofiler ble verifisert ved on-chip Giemsa-farging, og resultatene viste de typiske ringformede og lobeformede kjernene til nøytrofiler ( figur 2A ) 25 .
