Chemotaxis, en process med riktad cellmigrering till löslig kemisk koncentrationsgradient, är kritiskt inblandad i många biologiska förfaranden inklusive immunsvar 1 , 2 , 3 , vävnadsutveckling 4 och cancermetastas 5 . Neutrofiler är den vanligaste vita blodkroppsuppsättningen och spelar viktiga roller för att möjliggöra kroppens medfödda värdförsvarsfunktioner, samt för att förmedla adaptiva immunsvar 6 , 7 . Neutrofiler är utrustade med högreglerade kemotaktiska maskiner vilket gör att dessa motila immunceller kan reagera på både patogen-härledda kemoattraktanter ( t.ex. fMLP) och värd-härledda kemoattraktanter ( t.ex. interleukin-8) genomkemotaxis 8 . Neutrofil migration och kemotaxi medlar olika fysiologiska problemOch sjukdomar som inflammation och cancer 1 , 9 . Således ger den exakta bedömningen av neutrofilkemotaxis en viktig funktionell avläsning för att studera neutrofilbiologin och de associerade sjukdomarna. Jämfört med de allmänt använda konventionella kemotaxisanalyserna (t ex transwellanalys 10 ) visar de mikrofluidiska anordningarna ett stort löfte för kvantitativ utvärdering av cellmigration och kemotax på grund av den exakt kontrollerade kemiska gradientgenereringen och miniatyriseringen 11 , 12 , 13 . Under de senaste två decennierna har olika mikrofluidiska anordningar utvecklats för att studera kemotaxis av olika biologiska celltyper, speciellt neutrofiler 11 . Betydande ansträngningar ägnades åt att karakterisera neutrofilmigration i spatiotemporalt komplexa che<html> Mical gradienter som konfigurerades i de mikrofluidiska anordningarna 14 , 15 . Intressanta strategier utvecklades också för att studera riktningsbeslutande av neutrofiler med användning av de mikrofluidiska enheterna 16. Förutom biologisk orienterad forskning har tillämpningarna av mikrofluidiska anordningar utökats till testkliniska prover för utvärdering av sjukdomar 17 , 18 , 19 . Användningen av många mikrofluidiska anordningar är emellertid begränsad till specialiserade forskningslaboratorier och kräver långvarig neutrofilisolering från stor volym blodprover. Därför har det varit en växande trend att utveckla integrerade mikrofluidiska enheter för snabb analys av neutrofilkemotaxier direkt från en helblodsfall 20 , 21 , 22 ,Ef &quot;&gt; 23 , 24 . Mot denna riktning utvecklades en all-on-chip-metod som integrerar den magnetiska negativa neutrofila reningen och den efterföljande kemotaxisanalysen på en enda mikrofluidisk anordning 25 . Denna all-on-chip-metod har följande nya egenskaper: 1) Till skillnad från tidigare strategier på chip som isolerar neutrofiler från blodet genom vidhäftningsbaserad cellinfångning eller cellstorleksbaserad filtrering 20 , 22 tillåter denna nya metod hög Renhet, magnetisk separation på chipet av neutrofilerna från små volymer av helblod, liksom kemotaxismätning vid kemoattraktantstimulering; 2) celldockningsstrukturen hjälper till att justera de ursprungliga positionerna hos neutrofilerna nära kemisk gradientkanal och tillåter enkel kemotaxanalys utan singelcellspårning; 3) integrationen av neutrofilisolering och kemotAxelanalysen på en enda mikrofluidisk anordning tillåter snabb analys till resultat av kemotaxisanalys på 25 minuter när det inte uppstår något avbrott mellan försöksstegen. Detta papper ger ett detaljerat protokoll för konstruktion, drift och data analysmetod för denna all-on-chip chemotaxis analys. Mikrofluidiska enheter används alltmer för att undersöka neutrofilmigration och kemotaxor på grund av deras fördelar i realtidsvisualisering, exakt kontroll av kemisk koncentrationsgradientgenerering och minskad reagens- och provförbrukning. Nyligen har mikrofluidiska forskare gjort en ökande insats för att utveckla integrerade och lättanvända mikrofluidiska kemotaxisanalyssystem, direkt från helblod. I denna riktning utvecklades den första all-on-chip-metoden för att integrera den magnetiska negativa reningen av neutrofiler och kemotaxisanalysen från små blodvolymprover. Denna nya metod medger ett snabbt prov-till-resultat neutrofilt kemotaxistest på 25 min. I det här dokumentet tillhandahåller vi detaljerad konstruktion, drift och data analysmetod för denna allt-på-chip kemotaxis analys med en diskussion om felsökningsstrategier, limiTationer och framtida riktningar. Representativa resultat av den neutrofila kemotaxisanalys som testar en definierad kemoattraktant, N- formyl-Met-Leu-Phe (fMLP) och sputum från en patient med kronisk obstruktiv lungsjukdom (COPD) med användning av denna all-on-chip-metod visas. Denna metod är tillämplig på många cellmigrationsrelaterade undersökningar och kliniska tillämpningar.

Neutrofiler väljs negativt från en droppe helblod direkt i den mikrofluidiska enheten. Renheten hos de isolerade neutrofilerna verifierades genom on-chip Giemsa-färgning och resultaten visade de typiska ringformade och lobeformade kärnorna av neutrofiler ( Figur 2A ) 25 .
