Alkoholhaltig leversjukdom (ALD), som orsakas av alkoholöverkonsumtion, är en viktig orsak till alkoholrelaterad sjuklighet och mortalitet. I USA innebär nästan hälften av leversjukdomsdöd alkohol 1 , och ALD ansvarar för nästan 1 av 3 levertransplantationer 2 . ALD har ett brett spektrum. Steatos, som kännetecknas av överflödig lipidackumulering i hepatocyter, förekommer i det tidiga skedet av tung dricks och är reversibel vid upphörande av alkoholanvändning. Under påverkan av genetiska och miljömässiga faktorer och fortsatt alkoholintag kan hepatisk steatos utvecklas till alkoholhepitit och slutligen cirros 3 . Studier som använder ALD-modellerna för gnagare har gett väsentliga insikter i sjukdomen, men de har begränsningar (ses i referens 3 ). Oral utfodring av en alkoholdiet orsakar bara steatos hos gnagare 4 , &lt;/ Sup&gt; 5 . Utveckling av inflammation och fibros kräver antingen en andra förolämpning 6 , 7 eller kronisk intragastrisk infusion, vilket är invasiv och tekniskt utmanande 8 , 9 . Zebrafish lever är funktionell och producerar viktiga enzymer för etanol metabolism med 4 dagarS post-fertilization (dpf) 13 , 16 , 17. Ethanol kan direkt tillsättas till vattnet och exponering för 2% etanol i 24 timmar är tillräcklig för att inducera leverstatos och fibrogena svar i zebrafisklarver 13 , 15 . Det har rapporterats att behandling med 2% etanol i 24 h resulterade i en vävnad etanolkoncentration av 80 mM i zebrafisklarver 13 . Andra har visat att larver tolererar denna koncentration och de leverfenotyper som ses i de behandlade djuren är specifika för etanolexponering 11 , 13 , 15 , 18 . Eftersom 80 mM är nästan dödligt hos människor 19 är det emellertid viktigt att utvärdera levern histologi hos den etanolbehandlade zebrafisken och detaRmine den fysiologiska relevansen för människor. Den snabba externa utvecklingen och translucensen av zebrafisk larver gör det möjligt att karakterisera alkoholens verkan i levern i realtid och i fasta prover. Emellertid kan konfokal avbildning av den fluorescerande transgena zebrafisken inte helt ersätta hematoxylin och Eosin (H &amp; E) -färgning vid studier av leverhistologi. Markering av alla levercellstyper samtidigt som transgena zebrafiskar kräver generering av individuella transgena linjer, var och en märkning av en levercellstyp med en unik fluorofor. Att introducera olika transgena bakgrunder i samma fisk kräver rasinG flera generationer, vilket är tidskrävande och dyrt. Ytterligare immunofluorescensfärgning behövs för att detektera extracellulära matriskomponenter. H &amp; E-färgning, å andra sidan, märker samtidigt alla levercellstyper och extracellulära matriskomponenter, vilket ger en överblick över levern 20 . histological analyses of acute alcoholic liver injury in zebrafish

Alkoholhaltig leversjukdom (ALD) avser leverskador på grund av akut eller kronisk alkoholmissbruk. Det är bland de främsta orsakerna till alkoholrelaterad sjuklighet och dödlighet och drabbar mer än 2 miljoner människor i USA. En bättre förståelse för cell- och molekylära mekanismer som ligger till grund för alkoholframkallad leverskada är avgörande för att utveckla effektiv behandling för ALD. Sebrafisklarver uppvisar leverstatos och fibrogenes efter bara 24 timmars exponering för 2% etanol, vilket gör dem användbara för studier av akut alkoholhaltig leverskada. Detta arbete beskriver förfarandet för akut etanolbehandling i zebrafisklarver och visar att det orsakar steatos och svullnad i de leverblodkärlen. Ett detaljerat protokoll för hematoxylin och Eosin (H &amp; E) färgning som är optimerad för den histologiska analysen av zebrafisk larvleveren beskrivs också. H &amp; E-färgning har flera unika fördelar gentemot immunofluorescens, eftersom det markerar allt livCeller och extracellulära komponenter samtidigt och kan lätt detektera leverskada, såsom steatos och fibros.
