Det muliggjør analyse av protein-DNA samt protein-RNA-interaksjoner og spesielt interaksjoner mellom tRNA og tRNA-bindende proteiner ved bruk av enten rensede komponenter eller komplekse blandinger av proteiner ( f.eks . Vi har anvendt gel-retardasjonsanalyser til studien av samspillet mellom renset tRNA (UUU) og TcdA, en N6-treonylkarbamoyladenosin (t6A) dehydratase, som cykliserer treonylkarbamoyl-sidekjeden festet til A37 i anticodonstamsløyfen (ASL) Av tRNA til syklisk t6 (ct 6A) 2 , 3 . Dette er en fordel i forhold til alternative biofysiske teknikker, som vanligvis forbruker større mengder prøve. Gel-retardasjonsanalysen har blitt mye brukt til studiet av transkripsjonsfaktorer. Mange forskjellige parametere påvirker resultatet av en gelforsinkelsesanalyse, slik som temperatur, kvalitet på proteinet og tRNA-prøven og bindingsstyrken.
