De muis heeft ontpopt als een belangrijke diermodel voor menselijke pancreatic kanker1,2. In de Ptf1acre / +; LSL-KrasG12D / + muismodel, de Kristen rat Sarcoom (K-Ras) oncogen is geactiveerd uitsluitend in de alvleesklier, resulterend in de inleiding van voorstadia laesies in de alvleesklier, bekend als de alvleesklier geest neoplasias (PanINs), dat de vooruitgang aan alvleesklier ductaal adenocarcinomas, kortweg PDACs3. Deze muis modelsysteem biedt een van de beste beschikbare dierlijke modellen voor menselijke alvleesklierkanker4,5, met het bijkomende voordeel dat de PanINs ontstaan binnen de eerste vijf maanden van het leven en vaak vooruitgang PDAC binnen een Eén jaar4,5, overwegende dat alvleesklierkanker vaakst bij de mens 60-70 jaar oud optreedt.
Extractie van de alvleesklier door dissectie van de Ptf1acre / +; LSL-KrasG12D / + muizen op verschillende leeftijden voorziet in gedetailleerde longitudinale histopathologisch onderzoek van de ontwikkeling van kanker in de alvleesklier, variërend van de vroegste stadia van de PanIN de progressie tot PDAC3,4 , 5. de alvleesklier oogsten op leeftijden variërend van vijf tot vijftien maanden kan ook worden gebruikt om te bereiden weefsel extracten te karakteriseren van mondiale veranderingen in de alvleesklier4 metabolisme die zich tijdens de overgang van gezond naar zieke weefsel voordoen 6,7.
Dit artikel presenteert een complete visuele gids voor de stappen die nodig zijn voor het uitvoeren van een muis alvleesklier extractie en worden richtlijnen gegeven voor de opslag van een alvleesklier voor verdere analyse. Deze handleiding zullen even waardevol zijn voor individuen uitvoeren van onderzoek naar andere alvleesklier ziekten, met inbegrip van type I diabetes, en met name nuttig voor studenten en onderzoekers nieuw voor onderzoek waarbij oogsten moeten van het gebruik van de alvleesklier van de muis dissectie voor metabole profilering of histologische analyses.

We hebben onderzocht de alvleesklier specifieke transcriptiefactor, 1a cre-recombinase; Lox-stop-lox-Kristen rat Sarcoom, glycine aan asparaginezuur op de 12 codon (Ptf1acre / +; LSL-KrasG12D / +) stam muis als een model van menselijke pancreatic kanker. Het doel van onze lopende onderzoeken is het identificeren van de roman metabole biomarkers van pancreatic kanker progressie. Wij hebben uitgevoerd metabole profilering van urine, ontlasting, bloed, en alvleesklier weefsel extracten, evenals histologische analyses van de alvleesklier om de progressie van kanker te regisseren. De alvleesklier muis is niet een welomschreven solide orgel als bij de mens, maar is veeleer een diffuus gedistribueerde weke delen, dat is niet gemakkelijk geïdentificeerd door personen onbekend met interne anatomie van de muis of door personen die weinig of geen ervaring uitvoeren hebben muis orgel dissecties. Het doel van dit artikel is bedoeld als een gedetailleerde stapsgewijze visuele demonstratie bij beginners in de verwijdering van de alvleesklier muis door dissectie. Dit artikel moet vooral waardevol voor studenten en onderzoekers nieuw onderzoek waarvoor het oogsten van de alvleesklier muis door dissectie voor metabole profilering of histologische analyses.

Figuur 1 toont een overzicht van de operationele omgeving en Figuur 2 toont de post operatie gebied. Terwijl deze instelling de minimale hoeveelheid materiaal biedt en enscenering, individuen verkiezen kunnen om te veranderen dit best aan individuele behoeften.
