Wereldwijd is trombose een belangrijke doodsoorzaak en morbiditeit, en bloedplaatjes spelen een centrale rol bij de ontwikkeling ervan. Dit werk beschrijft een methode voor de live-celafbeelding van bloedplaatjes degranulatie onder stroming. Over het algemeen wordt aangenomen dat wanneer de bloedplaatjes geactiveerd worden alle korrelvormige inhoud direct in oplossing worden gebracht. De bijbehorende resultaten suggereren dat dit niet noodzakelijkerwijs het geval is. Tijdens adhesie en degranulatie behouden bloedplaatje...

Plateletten zijn anucleate cellen die in de bloedstroom circuleren. Hun belangrijkste functie is om vaatbreuken op plaatsen van letsel te verzegelen en om bloedverlies te voorkomen. Op deze plaatsen van letsel worden subendotheliale collageenvezels blootgesteld en worden ze daarna gedekt door het multimere eiwit, von Willebrand factor (VWF). VWF interactieert met de bloedplaatjes in omloop in een mechanisme dat afhankelijk is van het glycoproteïne Ibα-IX-V complex op het celoppervlak 1 , waardoor de snelheid van de bloedplaatjes wordt vertraagd. Dit is vooral belangrijk bij hoge schuifsnelheden. De bloedplaatjes ondergaan vervolgens morfologische veranderingen terwijl ze activerende impulsen ontvangen van collageen. Dit leidt tot onomkeerbare verspreiding en uiteindelijk tot aggregatie van bloedplaatjes. Beide processen zijn afhankelijk van de afscheiding van de korrelinhoud om transplantatie van bloedplaatjes-bloedplaatjes te vergemakkelijken. Bl.a. bevat plaatjes a-granules fibrinogeen en VWF om de hechting van de bloedplaatjes te ondersteunen en te overbruggenBloedplaatjes samen op integrin-afhankelijke wijze. De bloedplaatjes dichte korrels bevatten anorganische verbindingen 2 , waaronder calcium en adenosine difosfaat (ADP), die bijdragen tot het versterken van de bloedplaatjesactivering. Bovendien bevatten bloedplaatjes mediators van (allergische) ontsteking 3 , complement-controlerende eiwitten 4 en angiogenese factoren 5 , 6 , die de vragen verhogen over of en hoe deze inhoud onder verschillende omstandigheden vrijkomen. Sinds de jaren tachtig is de studie van bloedplaatjesfunctie in stromingsmodellen waardevol geweest voor het onderzoek van trombotische mechanismen 7 . Sindsdien is veel technische vooruitgang geboekt en stromingsmodellen die fibrinvorming bevatten, worden momenteel ontwikkeld om het hemostatische potentieel van therapeutische bloedplaatjesconcentraten ex vivo 8 te bepalen of omOnderzoek de invloed van verstoringen in schuifsnelheden op trombusmorfologie 9 . De verschillen in de moleculaire en celbiologische mechanismen die stabiele hechting en fysiologische trombusvorming (hemostase) vormen tegen de pathologische trombusvorming (trombose) kunnen zeer subtiel zijn en motiveren de ontwikkeling van stromingsmodellen die de real-time visualisatie van deze subcellulaire processen. Een voorbeeld van een proces waarvoor een dergelijke opstelling waardevol zou zijn, is de (her) verdeling van intracellulair polyfosfaat en de aanwerving van stollingsfactoren om de tijdsafhankelijke invloed op dit op fibrine-ultrastructuur 10 te ontdekken. Studies zijn vaak beperkt tot eindpuntanalyses. Het hoofddoel van de beschreven methode is het real-time visuele onderzoek mogelijk maken van dynamische subcellulaire processen die plaatsvinden tijdens de bloedplaatjesactivering onder stroming.

Bloedplaatjes zijn essentiële spelers in hemostase, de vorming van trombi om vasculaire breuken te verzegelen. Zij zijn ook betrokken bij trombose, de vorming van trombi die de vasculatuur en de verwondende organen afsluiten, met levensbedreigende gevolgen. Dit motivert wetenschappelijk onderzoek naar bloedplaatjesfunctie en de ontwikkeling van methoden om celbiologische processen te volgen als ze zich voordoen onder stromingsomstandigheden. Er zijn verschillende stromingsmodellen beschikbaar voor het bestuderen van bloedplaatjeshechting en aggregatie, twee belangrijke verschijnselen in de bloedplaatjesbiologie. Dit werk beschrijft een methode om real-time bloedplaatjes degranulatie te bestuderen tijdens de stroom tijdens de activatie. De methode maakt gebruik van een stroomkamer gekoppeld aan een spuitpompinstallatie die onder een brede veld, omgekeerde, LED-gebaseerde fluorescentiemicroscoop wordt geplaatst. De hier beschreven beschrijving zorgt voor de gelijktijdige excitatie van meerdere fluoroforen die worden afgeleverd door fluorescent gelabelde antilichamen of fluorescerenEnt kleurstoffen. Na live-cell imaging experimenten kunnen de dekselglazen verder worden verwerkt en geanalyseerd met behulp van statische microscopie ( dwz confocale microscopie of scan elektronenmicroscopie).
