Weltweit ist Thrombose eine führende Todesursache und Morbidität, und Thrombozyten spielen eine zentrale Rolle bei der Entwicklung. Diese Arbeit beschreibt ein Verfahren zur lebenden Zellenabbildung von Thrombozyten-Degranulation unter Strömung. Es wird allgemein angenommen, dass bei der Aktivierung von Blutplättchen alle körnigen Inhalte direkt in Lösung gelangen. Die begleitenden Ergebnisse deuten darauf hin, dass dies nicht unbedingt der Fall ist. Während der Adhäsion und Degranulation behalten die Plättchen...

Plättchen sind anucleare Zellen, die im Blutstrom zirkulieren. Ihre Hauptfunktion ist es, vaskuläre Brüche an Orten der Verletzung zu versiegeln und Blutverlust zu verhindern. An diesen Verletzungsorten werden subendotheliale Kollagenfasern exponiert und anschließend durch das multimere Protein, von Willebrand-Faktor (VWF) abgedeckt. VWF interagiert mit den Plättchen im Kreislauf in einem Mechanismus, der von dem Ibcoprotein Ibα-IX-V-Komplex auf der Zelloberfläche 1 abhängt, was die Geschwindigkeit der Thrombozyten verlangsamt. Dies ist besonders wichtig bei hohen Scherraten. Die Thrombozyten unterliegen anschließend morphologischen Veränderungen, während sie Aktivierungsimpulse aus Kollagen erhalten. Dies führt zu einer irreversiblen Ausbreitung und schließlich zu einer Blutplättchenaggregation. Beide Prozesse hängen von der Sekretion von Granulatinhalten ab, um das Plättchen-Plättchen-Übersprechen zu erleichtern. Unter anderem enthalten die Plättchen-α-Granulate Fibrinogen und VWF, um die Plättchenadhäsion zu unterstützen und zu überbrückenPlättchen zusammen in einer integrinabhängigen Weise. Die Thrombozyten-dichten Granulate enthalten anorganische Verbindungen 2 , einschließlich Calcium und Adenosindiphosphat (ADP), die zur Verstärkung der Thrombozytenaktivierung beitragen. Weiterhin enthalten die Blutplättchen Mediatoren von (allergischen) Entzündungen 3 , komplementkontrollierenden Proteinen 4 und Angiogenesefaktoren 5 , 6 , wobei die Frage aufgeworfen wird, ob und wie diese Inhalte unter verschiedenen Bedingungen unterschiedlich freigesetzt werden. Seit den 1980er Jahren ist die Untersuchung der Thrombozytenfunktion in Strömungsmodellen für die Untersuchung von thrombotischen Mechanismen wertvoll 7 . Seitdem wurden viel technischer Fortschritt gemacht, und Strömungsmodelle, die die Fibrinbildung einschließen, werden derzeit entwickelt, um das hämostatische Potential von therapeutischen Thrombozytenkonzentraten ex vivo 8 oder zu untersuchenUntersuchen den Einfluss von Störungen der Scherraten auf die Thrombusmorphologie 9 . Die Unterschiede in den molekularen und zellbiologischen Mechanismen, die eine stabile Adhäsion und physiologische Thrombusbildung (Hämostase) gegenüber der pathologischen Thrombusbildung (Thrombose) antreiben, können sehr subtil sein und die Entwicklung von Strömungsmodellen motivieren, die die Echtzeit-Visualisierung dieser Subzellulären ermöglichen Prozesse. Ein Beispiel für ein Verfahren, für das ein solches Setup wertvoll wäre, ist die (Re-) Verteilung von intrazellulärem Polyphosphat und die Rekrutierung von Gerinnungsfaktoren, um den zeitabhängigen Einfluss, den dies auf die Fibrin-Ultrastruktur 10 hat, aufzudecken. Studien sind oft auf Endpunktanalysen beschränkt. Das Hauptziel der beschriebenen Methode ist es, die visuelle visuelle Untersuchung von dynamischen subzellulären Prozessen zu ermöglichen, die während der Thrombozytenaktivierung unter Strömung stattfinden.

Blutplättchen sind wesentliche Akteure in der Hämostase, die Bildung von Thromben, um vaskuläre Brüche zu versiegeln. Sie sind auch an Thrombose beteiligt, die Bildung von Thromben, die das Gefäßsystem verschlingen und Organe verletzen, mit lebensbedrohlichen Konsequenzen. Dies motiviert die wissenschaftliche Forschung zur Thrombozytenfunktion und die Entwicklung von Methoden zur Verfolgung zellbiologischer Prozesse, wie sie unter Strömungsbedingungen auftreten. Für die Untersuchung der Thrombozytenadhäsion und -aggregation stehen eine Vielzahl von Strömungsmodellen zur Verfügung, zwei Schlüsselphänomene in der Thrombozytenbiologie. Diese Arbeit beschreibt eine Methode, um die Vollendung der Plättchendegranulation unter der Strömung während der Aktivierung zu untersuchen. Das Verfahren nutzt eine Strömungskammer, die mit einem Spritzenpumpenaufbau gekoppelt ist, der unter einem Weitfeld-, invertierten LED-basierten Fluoreszenzmikroskop angeordnet ist. Der hier beschriebene Aufbau ermöglicht die gleichzeitige Erregung von mehreren Fluorophoren, die von fluoreszenzmarkierten Antikörpern oder Fluoreszenzen abgegeben werdenEntfärbt. Nach Live-Zell-Imaging-Experimenten können die Deckgläser mit statischer Mikroskopie ( dh konfokale Mikroskopie oder Rasterelektronenmikroskopie) weiterverarbeitet und analysiert werden.
