live-cell imaging of platelet degranulation and secretion under flow

혈소판은 혈관 침범을 봉쇄하기위한 혈전 형성의 지혈에 필수적인 요소입니다. 또한 혈전증, 혈관을 막아 장기를 손상시키는 혈전 형성에 생명을 위협하는 결과를 초래합니다. 이것은 혈소판 기능에 대한 과학적 연구와 흐름 조건에서 일어날 때 세포 생물학적 과정을 추적하는 방법 개발의 동기를 부여합니다. 혈소판 접착 및 응집의 연구를 위해 다양한 유동 모델을 사용할 수 있는데, 혈소판 생물학의 두 가지 주요 현상입니다. 이 연구는 활성화 동안 혈류에서 실시간으로 혈소판 탈과립을 연구하는 방법을 설명합니다. 이 방법은 와이드 필드 역전 된 LED 기반 형광 현미경 아래에 놓인 주사기 펌프 셋업에 연결된 유동 챔버를 사용합니다. 여기에 설명 된 설정은 형광으로 표지 된 항체 또는 형광으로 전달되는 다중 형광 물질의 동시 여기를 허용합니다염료. 라이브 셀 이미징 실험 후 커버 유리는 정적 현미경 ( 즉, 공 초점 현미경 또는 주사 전자 현미경)을 사용하여 추가 처리 및 분석 될 수 있습니다.
