Globalt är trombos en ledande orsak till dödsfall och sjuklighet, och blodplättar spelar en central roll i utvecklingen. I det här arbetet beskrivs en metod för levande celler avbildning av blodplätt degranulering under flöde. Det antas generellt att när alla blodplättar aktiveras frigörs alla granulära innehåll direkt i lösning. De medföljande resultaten tyder på att detta inte nödvändigtvis är fallet. Blodplättar är anukleatceller som cirkulerar i blodflödet. Deras huvudsakliga funktion är att försegla vaskulära brott mot skadedjur och förhindra blodförlust. Vid dessa skadeställor blir subendoteliala kollagenfibrer exponerade och täcks därefter av det multimera proteinet, von Willebrand-faktorn (VWF). VWF interagerar med blodplättarna i omlopp i en mekanism som beror på glykoprotein Iba-IX-V-komplexet på cellytan 1 , vilket sänker hastigheten hos blodplättarna. Detta är särskilt viktigt vid höga skjuvhastigheter. Plättarna genomgår därefter morfologiska förändringar under mottagning av aktiveringsimpulser från kollagen. Detta leder till irreversibel spridning och så småningom till trombocytaggregation. Båda förfarandena beror på utsöndringen av granulainnehåll för att underlätta trombocyt-trombocytkroppsstörning. Bland annat innehåller blodplätts-a-granuler fibrinogen och VWF för att hjälpa trombocytadhesion och att överbryggaBlodplättar tillsammans på ett integrationsberoende sätt. De trombocyt-täta granulerna innehåller oorganiska föreningar 2 , innefattande kalcium och adenosindifosfat (ADP), vilket bidrar till att förstärka trombocytaktivering. Vidare innehåller trombocyter mediatorer av (allergisk) inflammation 3 , komplementskontrollande proteiner 4 och angiogenesfaktorer 5 , 6 , som lyfter frågan om huruvida och hur dessa innehåll är differentiellt frisläppta under varierande förhållanden. Sedan 1980-talet har studien av trombocytfunktion i flödesmodeller varit värdefull för undersökningen av trombotiska mekanismer 7 . Sedan dess har mycket tekniska framsteg gjorts och flödesmodeller som innefattar fibrinbildning utvecklas för närvarande för att analysera den hemostatiska potentialen av terapeutiska blodplättskoncentrat ex vivo 8 eller tillUndersöka påverkan av störningar i skjuvhastigheter på trombusmorfologi 9 . Skillnaderna i molekylära och cellbiologiska mekanismer som driver stabil vidhäftning och fysiologisk trombbildning (hemostas) mot patologisk trombbildning (trombos) kan vara mycket subtila och motivera utvecklingen av flödesmodeller som möjliggör realtidsvisualisering av dessa subcellulära processer. Ett exempel på ett förfarande för vilket en sådan inställning skulle vara värdefull är (åter) fördelningen av intracellulärt polyfosfat och rekryteringen av koagulationsfaktorer för att avslöja den tidsberoende påverkan som detta har på fibrin-ultrastruktur 10 . Studier är ofta begränsade till slutpunktsanalyser. live-cell imaging of platelet degranulation and secretion under flow

Blodplättar är viktiga aktörer i hemostas, bildandet av trombi för att försegla vaskulära brott. De är också involverade i trombos, bildandet av trombi som ockluderar kärlsystemet och skadade organ, med livshotande konsekvenser. Detta motiverar vetenskaplig forskning om trombocytfunktion och utveckling av metoder för att spåra cellbiologiska processer som de uppträder under flödesförhållanden. En mängd olika flödesmodeller finns tillgängliga för studier av trombocytadhesion och aggregering, två nyckelfenomen i blodplättsbiologi. I det här arbetet beskrivs en metod för att studera realtid i blodplättens degranulering under flöde under aktivering. Metoden använder sig av en flödeskammare kopplad till en sprutpumpuppsättning som placeras under ett brettfält, inverterat, LED-baserat fluorescensmikroskop. Inställningen som beskrivs här möjliggör samtidig excitering av flera fluoroforer som levereras av fluorescensmärkta antikroppar eller fluorescensEntfärger. Efter experiment med levande celler kan täckglasen bearbetas vidare och analyseras med statisk mikroskopi ( dvs. konfokal mikroskopi eller scanningelektronmikroskopi).
