Dünya çapında tromboz önde gelen ölüm ve morbidite nedenidir ve trombositler gelişiminde merkezi bir rol oynamaktadır. Bu çalışma akış altında trombosit degranülasyonunun canlı hücre görüntülemesi için bir yöntemi açıklamaktadır. Genellikle, trombositler harekete geçirildiğinde, tüm granül içeriği çözeltiye doğrudan salınır. Eşlik eden sonuçlar bunun mutlaka geçerli olmadığını göstermektedir. Trombositler kan akışında dolaşan anükleat hücrelerdir. Ana işlevi yaralanma yerlerinde vasküler yaraları kapatmak ve kan kaybını önlemektir. Bu yaralanma yerlerinde, subendotelyal kollajen lifleri maruz kalır ve daha sonra multimerik protein olan von Willebrand faktörü (VWF) ile kaplanır. VWF, trombosit hızını yavaşlatan hücre yüzeyi 1 üzerindeki glikoprotein Ibα-IX-V kompleksine bağlı olan bir mekanizma içinde dolaşımdaki trombositler ile etkileşime girer. Bu, yüksek makaslama hızlarında özellikle önemlidir. Trombositler daha sonra kollajenden aktive edici uyaranlar alırken morfolojik değişiklikler yaparlar. Kan trombositleri, hemostazda vasküler ihlalleri gidermek için trombüs oluşumu için önemli oyunculardır. Ayrıca tromboz, vaskülatürü tıkayan tromboz oluşumu ve organları yaralamaya, hayati tehlike oluşturan sonuçlarla ilgilenmektedirler. Yöntem, geniş alanlı, ters çevrilmiş, LED tabanlı floresan mikroskop altında yerleştirilen bir şırınga pompası kurulumuna bağlı bir akış odası kullanır. Burada açıklanan kurulum, flüoresan etiketli antikorlar veya fluoresc tarafından gönderilen birden fazla florofor eşzamanlı uyarılmasına olanak tanırBoyalar. Canlı hücre görüntüleme deneylerinden sonra kapak gözlükleri statik mikroskopi ( örn. Konfokal mikroskopi veya taramalı elektron mikroskobu) kullanılarak daha da işlenebilir ve analiz edilebilir.
