The protocol presented here constitutes a powerful biochemical tool for the study of the subsynaptic distribution of specific proteins within any brain region. However, there are some drawbacks inherent to the technique that deserve to be highlighted here. For instance, one of the main limitations is the relatively large amount of tissue needed to purify a reasonable amount of protein in order to perform the immunoblot analysis of all subsynaptic fractions. Die synaptische Übertragung beruht auf der physischen Integrität der Synapse, ein Konzept, das schon 1897 von Foster und Sherrington 1 vorgesehen war. Daher ist das Verständnis der Verteilung der Schlüsselneurotransmission-Komponenten ( z. B. Ionenkanäle, Rezeptoren usw. ) unerlässlich, um die synaptische Funktion sowohl bei normalen als auch bei pathologischen Zuständen aufzuklären. Die Elektronenmikroskopie (EM) hat enorm zur aktuellen ultrastrukturellen Vorstellung von Prototypen des zentralen Nervensystems (ZNS) beigetragen. Auf diese Weise hat EM die Unterschiede zwischen prä- und postsynaptischen Dichten, die durch eine Spalte mit einem ziemlich gleichmäßigen Abstand (~ 25 nm) getrennt sind, fein festgelegt. Interessanterweise weist die postsynaptische Apparatur eine relativ kontinuierliche, elektronendichte Verdickung unterhalb ihrer Plasmamembran, die sogenannte postsynaptische Dichte oder PSD 2 auf . Umgekehrt, bei der präsynaptischen Apparatur, eine bemerkenswerteE diskontinuierliches Cytomatrix-Netzwerk ist gerade unterhalb der Plasmamembran angeordnet, was für die Ausrichtung und das Andocken von synaptischen Vesikeln auf die aktive Zone der Plasmamembran wesentlich ist. Daher stellt EM den goldenen experimentellen Ansatz dar, um die Verteilung von Proteinen innerhalb strukturell konservierter ZNS-Synapsen zu untersuchen. Jedoch ist die Information, die durch Elektronenmikrographien bereitgestellt wird, statisch. In der Tat zeigen akkumulierende Beweise, dass in vivo Synapsen extrem dynamisch sind und so dramatische strukturelle Veränderungen bei anhaltender synaptischer Übertragung erleben. Darüber hinaus können sich die Morphologie und Zusammensetzung der Synapsen in verschiedenen ZNS-Regionen und bei der Entwicklung, Reifung, Alterung und der Entwicklung neuropathologischer Zustände ändern. Insgesamt stellt ein Protokoll zur Isolierung von Proteinen, die zu verschiedenen synaptischen Kompartimenten in physiologischen Zuständen gehören, ein wertvolles Werkzeug für eine umfassendere Untersuchung der synaptischen Funktion dar. <p cLass = &quot;jove_content&quot;&gt; Hier beschreiben wir diese Art von komplementärem experimentellen Ansatz, der die präparative biochemische Anreicherung der verschiedenen synaptischen Membranfächer ermöglicht - nämlich extra-, vor- und postsynaptische Membrandomänen. Dieses Membranfraktionierungsverfahren, das zuerst von Philips et al . (2001) 4 beruht auf einer pH-Verschiebung, die die in der prä- und postsynaptischen Apparatur auftretenden adhäsiven Wechselwirkungen schwächt. Zunächst ist es durch die Verwendung von milden Reinigungsmitteln bei pH 6,0 möglich, den Adhäsionsübergang zu erkennen, der die prä- und postsynaptische Apparatur hält und der von der extrasynaptischen Membrandomäne gehalten wird, die solubilisiert ist und somit aus den synaptischen Kontakten extrahiert werden kann. Anschließend schwächt die Erhöhung des pH-Werts von 6,0 auf 8,0 in Gegenwart von milden Reinigungsmitteln die Festigkeit des adhärenten Übergangs, der die präsynaptische aktive Zone fest an die postsynaptische Dichte gebunden hält. Daher ist das präsynaptische Fach soLubilisiert und kann von der postsynaptischen Dichte getrennt werden, die meistens erhalten bleibt, weil die Konzentration des verwendeten Waschmittels ihre Solubilisierung nicht begünstigt 4 . Interessanterweise kann die Fraktionierungseffizienz, eventuell höher als 90%, durch verschiedene subsynaptische Marker bestätigt werden: i ) synaptosomal assoziiertes Protein 25 (SNAP-25) aus der präsynaptischen aktiven Zone; Ii ) Synaptophysin aus der extrasynaptischen Fraktion ( dh außerhalb der aktiven Zone und einschließlich Mikrosomen); Und iii ) postsynaptisches Dichteprotein 95 (PSD-95), aus der postsynaptischen Dichte. Bemerkenswert ist, dass diese Hirnmembranfraktionierungsmethode erfolgreich eingesetzt wurde. Dementsprechend war es möglich, die subsynaptische Lokalisierung verschiedener Rezeptoren, wie alpha-Amino-3-hydroxy-5-methyl-4-isoxazolpropionsäure (AMPA) -Rezeptoren 5 , Adenosin-A 1 -Rezeptor (A 1 R) 6 präzise zu bestimmen ,Adenosin-A 2A -Rezeptor (A 2A R) 7 , Adenosintriphosphat (ATP) P2-Rezeptoren 8 , Nikotinacetylcholinrezeptor-Untereinheiten 9 und Parkinson-assoziierter Rezeptor GPR37 10 . Eine Reihe von Beschränkungen kann jedoch eine angemessene Beurteilung der synaptischen Verteilung eines bestimmten neuronalen Proteins behindern. brain membrane fractionation: an ex vivo approach to assess subsynaptic protein localization

Die Beurteilung der synaptischen Proteinzusammensetzung und -funktion stellt eine wichtige Herausforderung in der Neurowissenschaft dar. Allerdings ist es nicht einfach, die Neurotransmission, die in Synapsen auftritt, zu bewerten, da sie durch dynamische Protein-Protein-Wechselwirkungen und Phosphorylierungsereignisse stark reguliert wird. Dementsprechend ist es, wenn eine Methode verwendet wird, um synaptische Transmission zu studieren, ein wichtiges Ziel, diese vorübergehenden physiologischen Modifikationen zu bewahren. Hier stellen wir ein Hirnmembran-Fraktionierungsprotokoll vor, das ein robustes Verfahren zur Isolierung von Proteinen, die zu verschiedenen synaptischen Kompartimenten gehören, darstellt. Mit anderen Worten beschreibt das Protokoll eine biochemische Methodik zur Durchführung der Proteinanreicherung aus präsynaptischen, postsynaptischen und extrasynaptischen Kompartimenten. Zuerst werden Synaptosomen oder synaptische Terminals aus Neuronen gewonnen, die alle synaptischen Kompartimente mit einem diskontinuierlichen Saccharosegradienten enthalten. Bemerkenswert ist die Qualität dieser ersten synaptischen Membranvorbereitung cRituell Anschließend wird die Isolierung der verschiedenen subsynaptischen Kompartimente mit leichter Solubilisierung unter Verwendung von milden Reinigungsmitteln bei unterschiedlichen pH-Bedingungen erreicht. Dies ermöglicht eine Trennung durch Gradienten- und Isopycnic-Zentrifugationen. Schließlich wird die Proteinanreicherung an den verschiedenen subsynaptischen Kompartimenten ( dh vor-, post- und extrasynaptischen Membranfraktionen) mittels Immunoblotanalyse mittels gut charakterisierter synaptischer Proteinmarker ( dh SNAP-25, PSD-95 und Synaptophysin, ), So dass eine direkte Bewertung der synaptischen Verteilung eines bestimmten neuronalen Proteins ermöglicht wird.

Die beschriebene Methodik wurde weitgehend für die subsynaptische Analyse von neuronalen Proteinen im Allgemeinen und für die Isolierung und biochemische Charakterisierung der synaptischen Rezeptoren 5 , 6 , 7 , 8 , 9 insbesondere verwendet.
