הפיתוח של קהילות חיידקים תלוי בשילוב של גורמים דטרמיניסטיים וסטוכסטיים מורכבים שיכולים לשנות באופן דרמטי את ההתפלגות המרחבית ואת פעילותם של חברי הקהילה. פיתחנו פלטפורמת מערך microell שניתן להשתמש בה במהירות להרכיב ולעקוב אחר אלפי קהילות חיידקים במקביל. פרוטוקול זה מדגיש את השירות של הפלטפורמה ומתאר את השימוש שלה עבור ניטור אופטי של פיתוח של קהילות פשוטות, שתי חברות בתוך אנסמבל של מערכים בתוך הפלטפורמה. הדגמה זו משתמשת בשני מוטציות של Pseudomonas aeruginosa , חלק מסדרה של מוטציות שפותחו כדי לחקור פתוגניות של סוג VI. הוספת כרומוזומלית של גנים mCherry או GFP או להקל על הביטוי המכונן של חלבונים ניאון עם אורכי גל פליטה שונים שניתן להשתמש בהם כדי לפקח על שפע הקהילה ואת המיקום בתוך microwell. פרוטוקול זה מתאר מת &#39;מפורטד להרכבת תערובות של חיידקים לתוך הבארות של המערך ושימוש זמן לשגות הדמיה פלואורסציה ניתוח כמותי התמונה כדי למדוד את הצמיחה היחסית של כל אוכלוסייה חבר לאורך זמן. זריעה והרכבה של פלטפורמת microwell, ההליכים הדמיה הכרחי לניתוח כמותי של קהילות חיידקים בתוך המערך, ואת השיטות שניתן להשתמש בהם כדי לחשוף אינטראקציות בין מינים באזור מינים כל דנו.
