Veebesmettingen zijn een belangrijk probleem voor de veeindustrie in tropische en subtropische gebieden van de wereld, met de meest voorkomende methode van controle afhankelijk van het gebruik van acariciden 1 , 4 . Bm86 werd eerder geïdentificeerd binnen het epitheelvlak van de tikgut als beschermend antigeen tegen R. microplus infestatie 10 , met beperkt succes als een vaccinestrategie door Bm86 geografische sequentievariatie ...

Rhipicephalus microplus , de veestok, is de meest significante ectoparasiet in termen van economische impact op de veeindustrie van tropische en subtropische regio&#39;s, aangezien het vectoren boviene tik koorts (babesiosis), anaplasmose en paardenpiroplasmosis 1 , 2 , 3 , 4 is . Er zijn inspanningen gedaan voor veestokcontrole om het schadelijke effect te verminderen, maar conventionele methoden zoals het gebruik van chemische acariciden hebben impliciete nadelen, zoals de aanwezigheid van chemische residuen in melk en vlees, en de toename van de prevalentie van chemisch resistente tegels 5 , 6 , 7 . Bijgevolg is de ontwikkeling van alternatieve methoden voor tikbesturing onderzocht, zoals het gebruik van natuurlijk resistentiebeest, biologische bestrijding (biopesticiden) en vaccinInen 4 , 5 , 6 , 7 , 8 , 9 . In het nastreven van eiwitten die kunnen worden gebruikt als vaccin kandidaten, is het huidige onderzoek gericht op de tikgut. De midgutmuur is gebouwd uit een enkele laag epitheliale cellen die op een dunne basale lamina rusten, waarbij de buitenkant van de basale lamina een netwerk van spieren vormt. Licht- en elektronmicroscoopwaarnemingen wijzen erop dat de midgut bestaat uit drie typen cellen: reserve (ongedifferentieerd), secretie en spijsvertering. Het aantal celtypes varieert aanzienlijk afhankelijk van de fysiologische fase. Secretory en spijsverteringscellen zijn afkomstig van reservecellen 18 , 19 , 20 . De constructie van cDNA bibliothekenOm de samenstelling van de tikgoot te onderzoeken heeft geleid tot de identificatie van antigeen eiwitten, zoals Bm86, als potentiële vaccin kandidaten 2 , 3 , 4 . Het glycoproteïne Bm86 is gelokaliseerd op het oppervlak van bosluiscellen en veroorzaakt een beschermende immuunrespons tegen de veekenteken ( R. microplus ) bij gevaccineerde vee. Anti-Bm86 IgG&#39;s die worden geproduceerd door de geïmmuniseerde gastheer worden ingehaald door de tik, herken dit antigeen op het oppervlak van tikgutcellen, en versteur vervolgens de tikweefselfunctie en integriteit. Vaccins op basis van Bm86 antigenen hebben effectieve controle van R. microplus en Rhipicephalus annulatus getoond , door het aantal, gewicht en voortplantingscapaciteit van ingewikkelde vrouwtjes te verminderen, wat resulteert in een verminderde larve-besmetting in daaropvolgende tikgeneraties 4 . Bm86-gebaseerde vaccins zijn echter niet effectief tegen alle tikstadia en hebbenHeeft onbevredigende effectiviteit aangetoond tegen sommige geografische stammen van R. microplus , waardoor de rundvlees- en zuivelindustrie deze vaccins 2 , 4 slecht heeft aangenomen. Het vermogen om epitheelcellen uit de tikgut te isoleren is een belangrijke innovatie die de voortgang van het onderzoek mogelijk maakt om eiwitmembraansamenstelling te bepalen, waaronder morfologie en fysiologie onder verschillende omgevingsomstandigheden. De hier beschreven werkwijze maakt gebruik van het chelaatmiddel ethylendiaminetetraazijnzuur (EDTA) en het reductiemiddel tris (2-carboxyethyl) fosfine (TCEP) om het epithelium uit zijn subepitheelondersteunende weefsels 10 te ontlasten. Het epithelium wordt hersteld na mechanische verstoring van de weefsels door schudden, gevolgd door discontinuous gradient centrifugatie in Percoll. Dit papier beschrijft een haalbare en nieuwe techniek voor het isoleren van tikgut epiTheliale cellen. Biotinyleerde celoppervlakte-eiwitten, geïsoleerd uit het oppervlak van deze epitheliale cellen kunnen vervolgens worden geanalyseerd in downstream toepassingen, zoals FACS en / of LC-MS / MS-analyse.
