De fysieke eigenschappen van water en waterige mengsels in hun verschillende fasen zijn van fundamenteel belang en zijn belangrijk voor in vivo en in vitro begrip van biologische systemen. In de hedendaagse cryobiologie en biologische cryopreservering zijn de glazen of amorfe fasen van waterige cryoprotectantmengsels van bijzonder belang 1 , 2 . Nucleatie en groei van ijskristallen kunnen cellen en weefsels verstoren, en eiwit denaturatie en aggregatie bevorderen, zodat cryopreserverprotocollen die het oplosmiddel vertonen zijn steeds populairder geworden. Bij biomoleculaire kristallografie verstoort de kristallisatie van oplosmiddel in de kanalen tussen biomoleculen kristalroosters en verlaagt diffractie-eigenschappen. Vitrificatie wordt bereikt door middel van een combinatie van snelle koeling, uitdroging en toevoeging van cryoprotectieve opgeloste stoffen, zoals glycerol, ethyleenglycol, polyethyleenglycolen (PEG&#39;s),Alcoholen en zouten. Vitrificatie beperkt ijskristallisatie en -groei, maar elimineert niet alle koelverwante steekproeven. Het genereren van grote monsters uit een groot aantal kleine druppels op een manier die de leegtevolumes minimaliseert - een belangrijke bron van fout in de vorige metingen van de glasvezeldichtheid - is niet triviaal. Voor waterige mengsels kan differentiële verdamping van oplosmiddelcomponenten tijdens aerosolisatie en afzetting in een vacuüm leiden totAanzienlijke onzekerheden in gedeponeerde concentraties. We hebben een methode ontwikkeld op basis van cryoflotatie, waarmee nauwkeurige dichtheidsbepaling van waterige mengsels met behulp van individuele druppels zo klein mogelijk is als 50 pL 10 . Deze druppels kunnen snel afgekoeld worden, terwijl hun oorspronkelijke concentraties behouden worden, en hun cryogene temperatuurstoestand (verglaasd of kristallijn) kan worden beoordeeld aan de hand van een eenvoudige visuele analyse die correleert met röntgendiffractiemetingen. Deze methode is brede toepasbaar op waterige en niet-waterige mengsels en kan uitgebreid worden naar een verscheidenheid aan biologische monsters, waaronder cellen ( bijv . measuring the densities of aqueous glasses at cryogenic temperatures

Wij tonen een methode voor het bepalen van de cryogene temperatuurdichtheid van de vitreousfase van waterige mengsels en andere monsters die snelle koeling vereisen, om de gewenste cryogene temperatuurfase te bereiden. Metingen op waterige cryoprotectantmengsels zorgen voor inzicht in cryoprotectantwerking, en bieden kwantitatieve data om thermische samentrekking bij biologische cryopreservering te vergemakkelijken.
