Recent bewijsmateriaal heeft het begrip van de rollen van de vele regulerende mechanismen die na mRNA synthese optreden veranderd. Inderdaad kunnen translatie-, post-transcriptie- en proteolytische processen het eiwit overvloed en functie regelen. Het dogma - dat zegt dat mRNA concentraties proxies zijn aan die van de bijbehorende eiwitten, in het veronderstellen dat transcriptieniveaus de belangrijkste determinant van eiwit overvloed zijn - is gedeeltelijk herzien. In het begin voorspellen transcriptieniveaus slechts gedeeltelijk eiwit overvloed, wat suggereert dat posttranscriptiegebeurtenissen Voorkomen dat de eiwitten in cellen 1 , 2 gereguleerd worden . Het vastleggen van de complexiteit vanHet proteome vertegenwoordigt een technologische uitdaging. De huidige proteomische instrumenten bieden de mogelijkheid om op grote schaal analyses uit te voeren voor de identificatie, kwantificering en validatie van gewijzigde eiwitniveaus. Daarnaast heeft de introductie van fractionerings- en verrijkingstechnieken, die gericht zijn op het vermijden van de interferentie veroorzaakt door de meest overvloedige eiwitten, ook verbeterde eiwitidentificatie door de minste overvloedige eiwitten te omvatten. Tenslotte is proteomics aangevuld met de analyse van post-translational modificaties, die zich als belangrijke modulatoren van eiwitfunctie voordoen. De monstervoorbereiding en het eiwitherstel in de biologische monsters onder analyse blijven echter de beperkende stappen in de proteomische workflow en vergroten het potentieel voor mogelijke valkuilen 5 . Inderdaad, in de meeste moleculaire biologie technieken die moeten worden geoptimaliseerd, zijn de eerste stappen tissue homogenizatIon- en cellysis, in het bijzonder tijdens de analyse van eiwitten met weinig overvloed waarvoor geen amplificatiemethoden bestaan. Bovendien kan de chemische aard van eiwitten hun eigen herstel beïnvloeden. Bijvoorbeeld, de analyse van zeer hydrofobe eiwitten is zeer uitdagend omdat ze gemakkelijk neerslaan tijdens isoelectrische focus, terwijl transmembraan eiwitten bijna onoplosbaar zijn (in Referentie 5 onderzocht). Bovendien vormt de variatie van de weefselsamenstelling een belangrijke barrière voor het ontwikkelen van een universele extractiemethode. Ten slotte, omdat bijna alle klinische exemplaren van beperkte hoeveelheid zijn, is het essentieel om eiwitbereiding mogelijk te maken met maximaal herstel en reproduceerbaarheid van minimale monsterhoeveelheden 6 . Dit werk beschrijft een geoptimaliseerd protocol voor extractie van eiwitten uit de normale menselijke hart mitralisklep, die een zeer uitdagend monster voor proteomische analyse vertegenwoordigt. De normale mitrale klep is een compLex structuur tussen het linker atrium en de linker hartslag van het hart ( figuur 1 ). Het speelt een belangrijke rol in de controle van de bloedstroom van het atrium naar de ventrikel, het voorkomen van terugstroom en het verzekeren van de juiste zuurstofvoorziening aan het hele lichaam, waardoor een adequate hartuitgang wordt behouden. Het wordt echter vaak beschouwd als een &quot;inactief&quot; weefsel, met een lage cellulariteit en weinig componenten, voornamelijk in de extracellulaire matrix. Dit komt doordat de inwendige valvulaire interstitiële cellen (VIC&#39;s) onder normale omstandigheden een stil fenotype met een biosynthese van lage proteïnen 7 voordoen. Er is echter aangetoond dat in een pathologische toestand het aantal VICs in de spongiosa toeneemt en hun proteïnesynthese geactiveerd wordt, samen met andere functionele en fenotypische veranderingen 8 . Daarom is het niet verwonderlijk dat de minimale gegevens die beschikbaar zijn inDe literatuur richt zich op de analyse van pathologische mitrale kleppen 9 , 10 , waarin het verhoogde aantal geactiveerde VIC&#39;s het relatief hoge aantal geïdentificeerde eiwitten kan verklaren. Ten slotte kan het onderhavige protocol dienen om het begrip van de pathogene mechanismen die verantwoordelijk zijn voor mitrale klepziekten te ontwikkelen door middel van het bestuderen van mitrale klep eiwit componenten. Inderdaad, een beter begrip van de onderliggende pathologische processen kan bijdragen tot het verbeteren van het klinisch beheer van klepziekten, waarvan de huidige indicaties voor interventie grotendeels op hemodynamische overwegingen zijn gebaseerd.

optimized protocol for the extraction of proteins from the human mitral valve

Analyse van het cellulaire proteome kan bijdragen tot het verhelderen van de moleculaire mechanismen die aanliggende ziekten zijn door de ontwikkeling van technologieën die de grootschalige identificatie en kwantificering van de eiwitten in complexe biologische systemen mogelijk maken. De kennis die wordt verkregen uit een proteomische benadering kan mogelijk leiden tot een Beter inzicht hebben in de pathogene mechanismen die onderliggende ziekten liggen, waardoor de diagnose van nieuwe diagnostische en prognostische ziekte markers mogelijk wordt gemaakt, en hopelijk van therapeutische doelen. Echter, de cardiale mitrale klep vertegenwoordigt een zeer uitdagend monster voor proteomische analyse vanwege de lage cellulariteit in proteoglycan en collageenverrijkte extracellulaire matrix. Dit maakt het uitdagend om eiwitten te extraheren voor een globale proteomische analyse. Dit werk beschrijft een protocol dat compatibel is met latere eiwitanalyse, zoals kwantitatieve proteomics en immunoblotting. Dit kan voor de correlatie van gegevens zorgenG eiwit expressie met gegevens over kwantitatieve mRNA expressie en niet-kwantitatieve immunohistochemische analyse. Inderdaad zullen deze benaderingen leiden tot een uitgebreider begrip van de moleculaire mechanismen die onderliggende ziekten liggen, van mRNA tot post-translatie eiwitmodificatie. Zo kan deze methode relevant zijn voor onderzoekers die geïnteresseerd zijn in de studie van hartklep-fysiopathologie.
