유전자 조작 마우스의 사용은 생체 내 및 병리학 적 생체 내 과정 모두에 대한 연구에 크게 기여했다. 수정 된 난 모세포 내로의 DNA 발현 구조물의 전 핵 주입은 과발현을위한 형질 전환 쥐를 생성하는 가장 보편적 인 기술로 남아있다. 유전자 표적화를위한 CRISPR 기술이 도입됨에 따라 수정 된 난 모세포에 전핵을 주입하여 knockout과 knockin 마우스를 생성 할 수있게되었다. 이 연구는 유전자 타겟팅을위한 사출 용 DNA의 준비와 CRISPR 가이드의 생성을 기술하고 있으며 특히 품질 관리에 중점을두고 있습니다. 잠재적 인 창시자를 확인하는 데 필요한 유전형 분석 절차가 중요합니다.
