Eerder, Hofmann en collega's vinden dat dramatische morfologische veranderingen werden waargenomen in de REF52 cellen na de microinjection van de gratis PKA-19. Chemisch gemodificeerde eiwitten die photoactivated (bijvoorbeeld PKA gekooide eiwitten worden kunnen) zijn ontwikkeld als een opkomende veld in de chemische biologie te niet-gebeurt manipuleren intercellulaire biochemische processen1,2 ,3. Met behulp van licht, zoals een stimulans uitstekende Spatio resolutie biedt bij het activeren van deze gekooide eiwitten. UV-licht kan echter leiden tot ongewenste morfologische veranderingen, apoptosis en DNA schade aan cellen4,5. Vandaar, recente ontwikkelingen in het ontwerp van photocaging groepen richten over het inschakelen van photocleavage bij langere golflengte of twee-foton excitatie om fototoxiciteit, evenals over het verhogen van de diep-weefsel penetratie6,7. Kooien groepen die reageren op langere golflengte waardoor ons om te kiezen van de geschikte uncaging golflengten (d.w.z., kanalen) selectief activeren bioeffectors wanneer twee of meer caging groepen zijn huidige7. Gezien deze handige functies, is ontwikkeling van nieuwe rood-licht photocaging groepen zeer belangrijk stroomopwaartse werk in fotochemische methodologieën voor biologische studies, variërend van het sonderen van de mechanismen van de reacties op het beheersen van cellulaire activiteiten8. Niettemin, een twee-foton caging groep is normaal als gevolg van de structuur van de gesmolten aromatische ring te hydrophobic, en een caging groep van zichtbaar licht is normaal organometaal, met aromatische liganden. Deze hydrofobe/aromatische eigenschap is niet geschikt als de bioeffector is een eiwit of enzym, zoals het denatureert van de site van de activering van het enzym/eiwit en verlies van functie, veroorzaakt zelfs als de vervoeging en fotolyse nog steeds op de chemische niveau2 werkt ,9.
UCNPs zijn effectieve omvormers die het NIR excitatie licht omzetten in UV. Deze unieke en fascinerende eigenschap van UCNPs biedt realistische oplossingen voor de uitdagingen in verband met photoactivation en geactiveerd gecontroleerde afgifte van kleine molecules, met inbegrip van foliumzuur10, cisplatine derivaten11 , DNA/siRNA12, copolymeer blaasjes13en holle deeltjes14. Echter tot de beste van onze kennis, is de UCNP-bijgewoonde photoactivation van enzymen of eiwitten niet getest tot nu toe. Omdat er geen succesvolle geldt voor het gebruik van rood licht of NIR photoactive een enzym, werden we gevraagd uit te voeren van de NIR-geactiveerd activering van een eiwit/enzym constructie bestaat uit chemisch gewijzigde gekooide enzym complexen met een silica-bekleed, lanthanide-doped UCNP15. In dit onderzoek was de UCNP met een snel reagerende signaaltransductie kinase in de vorm van gekooide PKA vervoegd. PKA regelt glycogeen-synthese en cytoskeletal verordening die op externe prikkels via cyclisch adenosine fosfaat (cAMP) verordening in het cytosol16 reageert. We studeerde de haalbaarheid van enzymactivering in temporele en ruimtelijke manieren in een cellulaire experiment na NIR bestraling. Dit UCNP-bijgewoonde photoactivation platform is een nieuwe methode om photoactivate een enzym met behulp van NIR en vermijdt de ongewenste signaaltransductie reactie van cellen veroorzaakt door conventionele UV-bestraling-2,4.
Het is zeer moeilijk te translocate grote bioeffectors (bijvoorbeeld eiwitten) tussen de celmembranen controle van cellulaire activiteit. Hoewel deeltje-geïmmobiliseerd eiwit gemakkelijker translocate via endocytose in het cytosol wellicht, kan endocytose worden beschadigd of gedegradeerd via endosomal entrapment en de daaruit voortvloeiende lysosomale afbraak2,4. Misschien zelfs als de gekooide proteïne nog steeds functioneel na membraan translocatie is, de translocated bedragen niet genoeg om het activeren van de cellulaire reactie2,17. In schril contrast is microinjection een directe en kwantitatieve benadering leveren grote bioeffectors naar het cytoplasma van de cel. Bovendien vereist de bioeffector UCNP-geïmmobiliseerd upconverted licht te worden geactiveerd. De optische instrumenten vereist daarom verdere wijziging te meten, visualiseren, en gebruik maken van het licht upconversion. In dit werk, zal de levering van een gekooide PKA-UCNP-complex naar een cel met behulp van microinjection en de volgende essentiële spectroscopie en microscopie wijzigingen voor NIR photoactivation in detail worden beschreven.

an integrated system to remotely trigger intracellular signal transduction by upconversion nanoparticle-mediated kinase photoactivation

Upconversion nanoparticle (UCNP)-gemedieerde photoactivation is een nieuwe aanpak om op afstand controle bioeffectors met veel minder fototoxiciteit en diepere weefsels penetratie. De bestaande instrumenten op de markt is echter niet gemakkelijk compatibel met upconversion toepassing. Wijzigen van de commercieel beschikbare instrument is daarom essentieel voor dit onderzoek. In deze paper illustreren we eerst de wijzigingen in een conventionele fluorimeter en fluorescentie Microscoop te laten compatibel voor foton upconversion experimenten. Vervolgens beschrijven we de synthese van een nabij-infrarood (NIR)-geactiveerd gekooide proteïne kinase A katalytische subeenheid (PKA) geïmmobiliseerd op een UCNP complex. Parameters voor microinjection en NIR photoactivation procedures worden ook gemeld. Nadat de gekooide PKA-UCNP is microinjected in REF52 fibroblast cellen, activeert de bestraling van het NIR, dat aanzienlijk superieur aan conventionele UV-bestraling is, efficiënt de PKA signaaltransductie traject in levende cellen. Bovendien bevestigen de positieve en negatieve controle experimenten dat de PKA-geïnduceerde traject leidt tot het uiteenvallen van stress vezels specifiek wordt geactiveerd door NIR bestraling. Het gebruik van gemodificeerde eiwitten UCNP biedt dus een innovatieve benadering van op afstand bedienen licht-gemoduleerd cellulaire experimenten, waarin directe blootstelling aan UV-licht vermeden worden moet.

Het ontwerp van de gekooide enzym-UCNP constructie wordt geïllustreerd in Figuur 1. Het enzym PKA was eerst reageerden met 2-nitrobenzyl bromide (en) voor het genereren van een inactieve gekooide PKA, en het werd vervolgens via een elektrostatisch proces geïmmobiliseerd op het oppervlak van UCNP. UCNPs de upconverted licht uitstralen en dus photolytically het klieven van de o-nitrobenzyl groepen op Cys 199 en Cys 343, het genereren van de geactiveerde PKA. ...
