Het gebruik van volgende generatie sequencing technieken om globale DNA-methylering profielen te analyseren is in de afgelopen jaren steeds populairder geworden. Genoom-brede methyleringsbepalingen, met inbegrip van microarray- en niet-microarray gebaseerde methoden, werden ontwikkeld op basis van het volgende: de bisulfietomzetting van genomisch DNA, methylatiegevoelige restrictie-enzymverdigingen en de immunoprecipitatie van gemethyleerd DNA onder gebruikmaking van methyl-CpG-specifieke antilichamen . Aberrant DNA methylering is een van de kenmerken van leukemie en lymfomen, waaronder chronische lymfocytische leukemie (CLL). Vroeger hebben verschillende groepen, waaronder onze, de DNA-methyleringsprofielen van verschillende CLL-prognostische subgroepen en normale, gezonde B-celcontroles gekenmerkt door gebruik te maken van de bisulfietomzetting van genomisch DNA, gevolgd door micro-array-gebaseerde methoden of sequentiebepaling van het genoom-genoom 1 , 2 , 3 , &lt;Sup class = &quot;xref&quot;&gt; 4. Inmethyl-CpG-bindende domein (MBD) sequentiebepaling van volgende generatie (MBD-seq), is de verrijking van dubbelstrengs gefragmenteerd DNA afhankelijk van de mate van CpG-methylering. Deze methode kan de nadelen van de bisulfietomzettingsmethode overwinnen en kan ook genoom-brede dekking van CpG-methylering op een onbevooroordeelde en PCR-onafhankelijke wijze verschaffen. Bovendien kan MBD-seq, in tegenstelling tot bisulfietomzettingsgebaseerde microarraymethoden, worden gebruikt om de methyleringsstatus van herhalende elementen te analyseren, zoals langdurige kernelementen (LINE&#39;s), korte interversionele kernelementen (SINE&#39;s), lange terminale herhalingen (LTRS), Enz . In vergelijking met bisulfietomzettingsmethoden,Een MBD-seq protocol vereist een relatief grote hoeveelheid input DNA. In principe maakt deze methode gebruik van het vermogen van de MBD van de menselijke MBD2-eiwitinteractie met gemethyleerde CpG&#39;s om DNA te verrijken verrijkt met gemethyleerde CpG&#39;s, en dit wordt gevolgd door de high-throughput sequencing van gemethyleerd DNA.

De rol van lange noncoding RNAs (lncRNAs) bij kanker komt naar voren als gevolg van de groeiende interesse in het begrijpen van hun mechanistische functies tijdens de ontwikkeling van kanker en progressie. Desondanks is de globale epigenetische regulering van lncRNAs en repetitieve sequenties bij kanker niet goed onderzocht, vooral bij chronische lymfocytische leukemie (CLL). Deze studie richt zich op een unieke aanpak: de immunoprecipitatie-gebaseerde opname van dubbelstrengige, gemetileerde DNA-fragmenten met behulp van methyl-bindende domein (MBD) eiwitten, gevolgd door volgende generatie sequencing (MBD-seq). CLL patiëntenmonsters die behoren tot twee prognostische subgroepen (5 IGVH gemuteerde monsters + 5 IGVH niet gemuteerde monsters) werden in deze studie gebruikt. Analyse onthulde 5.800 hypermethylated en 12,570 hypomethylated CLL-specifieke differentieel gemethyleerde genen (cllDMGs) in vergelijking met normale gezonde controles. Het is belangrijk dat deze resultaten verschillende CLL-specifieke, differentieel gemethyleerde lncRNAs, opnieuw identificerenPetitieve elementen, en proteïne-coderende genen met potentiële prognostische waarde.
