En av biomarkørene som utforskes, er 3-nitrotyrosin (3-NT), et sluttstabil produkt dannet når reaktive nitrogenarter (RNS) interagerer med tyrosin, en katecholamin-neurotransmitter-forløper. Selv om 3-NT kan ha klinisk verdi som biomarkør for RNS in vivo, kan de betydelige endringene i tyrosins egenskaper og funksjoner påvirke tilsvarende proteiner og cellefunksjoner 1 , 2 negativt. Ny forskning har antydet at 3-NT kan spille en viktig rolle i inflammatoriske tilstander 3 , neurodegenerative lidelser 4 , 5 , kardiovaskulær sykdom 6 og diabetes 7 samt forhold knyttet til oksidativt stress. Men disse obseRvasjonene er basert på resultater fra metoder som mangler sensitivitet og / eller selektivitet 8 , 9 , 10 , 11 . De enorme 3-NT konsentrasjonsområder for de biologiske prøver som tidligere er rapportert i litteraturen, viser at alvorlige analytiske problemer er forbundet med disse analysene, og det er nødvendig med teknisk forbedring for å nøyaktig kvantifisere nivåene av 3-NT og verifisere sin rolle i patologien til disse tilstandene . Kvantifiseringen av gratis 3-NT i biologiske matriser gir en spesiell utfordring for menneske og instrument 8 , 9 , 10 , 11 . Gaskromatografi (GC) koblet MS / MS gir den beste følsomheten, men de uunnværlige prøve derivatiseringstrinnene er for kjedelige og tidkrevende for å være effektive for klinisk bruk 15 , 16 . LC-MS / MS krever ikke komplisert prøve derivatisering, noe som gjør det mer lovende alternativ. Likevel er det flere hindringer for å overvinne, slik som seNsitiviteten til LC-MS / MS-metodene som er rapportert i litteraturen, må forbedres for måling av lite rikelig 3-NT 7 , 17 , 18, og den relativt lange ventetiden må forkortes for applikasjoner med høy gjennomstrømning 12 , 13 , 17 , 19 . I tillegg, når man vurderer kliniske anvendelser, spiller den biologiske matrisen en betydelig rolle. Det bør være enkelt og rimelig å skaffe seg og ikke-invasiv hvis mulig 20 , 21 , 22 . Plasma, den tradisjonelt brukte prøven i litteraturen, er ikke en klinisk ønskelig matrise, så en metode som benytter urin som er ikke-invasiv og kostnadseffektiv, ble søkt. Flere forsøk på å utvikle relIable og spesifikke LC-MS / MS metodikker har blitt gjort ved bruk av urin 9 , 10 , 11 . Imidlertid har de alle manglet å være selektive, pålitelige eller effektive nok til klinisk bruk. Effektiviteten til den overveiende SPE ved bruk av tradisjonell reversertfasepatron (C18 type) som prøveopprydding for 3-NT-analysen, er blitt undersøkt, og en sekventiell SPE med sterk kationutveksling (SCX) og reversert fase C18-OH er foreslått 6 , 7 , 19 . En nylig utviklet LC-MS / MS-metode benyttet en multi-trinns renseprosess med manuell C18 SPE, preparativ høytrykksvæskekromatografi (HPLC) og online SPE for analyse av 3-NT 23 . Selv om denne metoden var sensitiv nok til kliniske formål, med en LLOQ på 0,041 nM, var oppryddingsprosessen intensiv og kjedelig og requiRød 3 ml urin, noe som begrenser muligheten for høy gjennomstrømning. En molekylært påtrykt polymer ble anvendt som SPE-sorbenten for å forbedre effektiviteten av oppryddingsprosessen 14 , men den resulterende LLOQ (0,7 ug / ml) var ikke lav nok til kliniske prøver. En annen metode krevde todimensjonal (2D) LC-MS / MS og immunaffinitetskromatografi for prøveopprydding for å oppnå en gjenkjenningsgrense (LOD) på 0,022 nM 24 . Mens alle disse metodene har gjort fremskritt i vurderingen av 3-NT, har ingen oppnådd følsomheten, påliteligheten og effektiviteten som er nødvendig for kliniske anvendelser. For å undersøke patologien til gratis 3-NT og dets rolle som en biomarkør av oksidativt stress i kliniske innstillinger, har vi utviklet en metodikk som er enkel, effektiv, nøyaktig og presis, og muliggjør kliniske applikasjoner med høy ytelse 25 . En miniaturisert blandet modus kation eksk(MCX) 96-brønns ekstraksjonsmikroplate ble implementert for å oppnå enkel og effektiv prøveopprensing og anrikning av 3-NT i en enkelt ekstraksjon omgåelse av ulempene sett i de eksisterende metodene som krever derivatisering, fordamping og 2D-LC. Væskekromatografi med 0,01% HCOOH som additiv i mobilfase ga et forbedret signalrespons med en rask syklus tid. Selektivitet ble ytterligere forbedret ved anvendelse av en mild NH4OAC elueringsoppløsning for selektiv eluering av 3-NT, og bruk av MRM overgang for både 3-NT og den interne standard (IS). Matrikseffekten ble kompensert ved å bruke en redusert mengde av en foretrukket 13 C-merket isotopisk IS for kvantifisering. Med fremkomsten av denne metoden vil forskere og klinikere kunne verifisere rollen som 3-NT i kliniske forhold og videre undersøke virkningen av oksidativt stress.

integration of miniaturized solid phase extraction and lc-ms/ms detection of 3-nitrotyrosine in human urine for clinical applications

Fri 3-nitrotyrosin (3-NT) har blitt brukt som en mulig biomarkør for oksidativt stress. Økte nivåer av 3-NT har blitt rapportert i mange forskjellige patologiske forhold. Eksisterende metoder mangler imidlertid tilstrekkelig sensitivitet og / eller spesifisitet som er nødvendig for å måle det lave endogene nivået av 3-NT pålitelig og er for tungt for kliniske anvendelser. SPE ved hjelp av en 96-brønnplate forenklet prosessen markant ved å kombinere prøveopprydding og analyse-anrikning uten kjedelige derivatisering og fordampningstrinn, Reduserer løsemiddelforbruk, avfallshåndtering, risiko for forurensning og total behandlingstid. Anvendelsen av 25 mM ammoniumacetat (NH4OAC) ved pH 9 som SPE elueringsoppløsningen vesentlig forbedret selektivitet. Massespektrometrisignalrespons ble forbedret gjennom justering av multiple reaksjonsovervåking (MRM) overganger. Bruk av 0,01% HCOOH som tilsetningsstoff på en pentafluorfenyl (PFP) kolonne (150 mm x 2,1, 3 um) forbedret signalresponsen en annen 2,5 ganger og forkortet total kjøretid til 7 min. En nedre grense for kvantifisering (LLOQ) på 10 pg / mL (0,044 nM) ble oppnådd, hvilket representerer en signifikant følsomhetsforbedring over de rapporterte analysene. Denne forenklede, raske, selektive og følsomme metoden tillater to plater av urinprøver (n = 192) å behandles i en 24 h tidsperiode. Figur 1 illustrerer at 3-NT er helt kromatografisk separert fra andre strukturelt lignende tyrosinanaloger under den optimaliserte LC-tilstanden, som eliminerer de sammenløpende interferensene på grunn av disse meget overdrevne forbindelser og følgelig forbedrer graden av assayselektivitet.
